Identification of common wheat and spelt by DNA markers

Abstract:

Pšenice špalda se v poslední době stává stále oblíbenější a žádanější vzhledem k lepšímu nutričnímu složení potravin ze špaldy oproti klasickým potravinám z pšenice seté. Obě tyto rostliny jsou jedny ze základních obilovin, pěstují se po celém světě a využívají se především v potravinářství a krmivářství. Vzhledem k častému falšování špaldových produktů prostřednictvím příměsí z pšenice seté roste zájem o jejich vzájemné odlišení. Jedním z možných nástrojů pro jejich identifikaci jsou metody molekulární biologie. Tímto směrem byla také zaměřena tato práce s názvem „Identifikace pšenice seté a špaldy DNA markery“. Testovaly a ověřovaly se DNA markery spojené s variabilitou v genu pro γ-gliadiny prostřednictvím PCR reakce a restrikčního štěpení (metoda RFLP). Bylo testováno celkem devět genotypů rodu pšenice a výsledky byly ověřovány na polyakrylamidové elektroforéze a pomocí bioinformatický nástrojů. Z výsledku vyplývá, že uvedený polymorfismus v genu pro γ-gliadiny lze použít jako identifikátor pro rozlišení špaldy a pšenice seté.

Abstract:

Spelt has recently become more popular and desirable due to the better nutritional composition of foods containing spelt, compared to traditional foods from common wheat. Both of these plants are some of the basic cereals, they are grown all over the world and are mainly used in the food and feed industries. Due to the frequent adulteration of spelt products through the use of common wheat admixtures, there is a growing interest in their differentiation. Molecular biology methods are one of the possible tools for identifying them. This work entitled “Identification of common wheat and spelt by DNA markers” was focused in this direction as well. DNA markers associated with variability in the γ-gliadin gene through PCR reaction and restriction cleavage (RFLP method) were tested and verified. Nine genotypes of the wheat genus were tested and the results were verified on polyacrylamide electrophoresis and by means of bioinformatics tools. The result shows that the polymorphism in the γ-gliadin gene can be used as an identifier for distinguishing spelt and common wheat.
