Bakteriální komponenty jako nanopřenašeče pro transport léčiv a jejich využití v péči o živočišné a lidské zdraví

Abstract:

Bakteriální rezistence na antimikrobiální látky patří v dnešní době mezi jednu z největších světových hrozeb. Z tohoto důvodu se také léčba infekcí, způsobených pato-genními bakteriemi, stala výzvou pro moderní medicínu. Proto jsou pro boj s bakteriální rezistencí vyvíjeny alternativy ke komerčním antibiotikům také z odvětví nanotechnolo-gie, využívající nanočástice s antimikrobiálním účinkem nebo pro transport antibiotik. Těchto nanomateriálů pro dopravu léčiv bylo navrženo velké množství a dnes je v tomto odvětví velký zájem i o využití bakteriálních komponent. V praktické části diplomové práce byly testovány bakteriální ferritiny; apoferritin izolovaný z bakterie Pyrococcus furiosus (PFU), jeho modifikované formy s delecí 15 (PFU C15) a 20 (PFU C20) aminokyselin a bakterioferritin (BFR), jako potenciální transportéry antibiotik. Nejdříve byla studována disociace ferritinů při různém pH (pH 1 – 12,5) a iontové síle (0 – 1000 mM) pomocí nativní polyakrylamidové gelové elektrofo-rézy. Výsledky prokázaly, že pro následnou enkapsulaci antibiotik je vhodný PFU, PFU C15 a BFR, na rozdíl od PFU C20, který byl rozložený na podjednotky za všech pod-mínek. Průtoková cytometrie odhalila, že bakteriální ferritiny jsou schopny interagovat s patogenními bakteriemi; buď přes povrchové receptory nebo inkorporací do bakteriál-ních buněk díky své nanovelikosti. Poté byly ferritiny PFU, PFU C15 a BFR využity pro enkapsulaci antibiotik; cefoxitinu, vankomycinu a tetracyklinu. Nejlepší výsledky byly docíleny pro PFU C15 s vysokou enkapsulační výtěžností a optimální velikostí pro tetracyklin (279.91 μg/mL, 13.1 nm) a cefoxitin (247.53 μg/mL, 14.8 nm), ale byl pozo-rován i výskyt větších ferritinových frakcí (200 – 500 nm). Enkapsulovaná antibiotika byla dále otestována na různých bakteriálních kmenech, a také na biofilmu tvořeným bakteriemi S. aureus. Enkapsulovaný cefoxitin neinhiboval bakteriální růst, pravděpo-dobně kvůli jeho degradaci během enkapsulace; a enkapsulovaný vankomycin neredu-koval vankomycin rezistentní buňky, nanočástice pravděpodobně nebyly schopny pře-konat obranné mechanizmy buněk. Enkapsulovaný tetracyklin měl stejný inhibiční efekt jako samotný tetracyklin na planktonní buňky i na biofilm tvořený bakteriemi S. aureus.

Abstract:

Nowadays bacterial resistance towards antimicrobials belong to the one of the biggest global threats in the world and infections caused by pathogenic bacteria are the challenge for modern medicine to treat them only via antibiotics. Many alternatives have been developed to fight with bacterial resistance, especially from the field of nanotech-nology, such as nanoparticles with intrinsic antimicrobial properties or nanocarriers for drug delivery. Various types of these nanocarriers have been designed and nowadays there is also an interest in the bacterial components employment.In experimental part of diploma thesis, the bacterial ferritin nanocages, namely Pyrococcus furiosus apoferritin (PFU), its modified versions with deletion of 15 amino acids (PFU C15) and 20 amino acids (PFU C20) and bacterioferritin (BFR), were tested as potential drug delivery nanocarriers. Their properties of assemble and disassemble were examined by using native polyacrylamide electrophoresis after exposure to differ-ent pH (1 – 12.5) and ionic strength (0 – 1000 mM). The results showed that PFU, PFU C15 and BFR are applicable for encapsulation of antibiotics. On the contrary, PFU C20 was dissociated under all conditions. Flow cytometry showed that bacterial ferritins are able to interact with pathogenic bacteria; either by binding to receptors or incorporating inside of bacteria due to its nanosize. Then PFU, PFU C15 and BFR were utilized for encapsulation of antibiotics; cefoxitin, vancomycin and tetracycline. The best results were obtained for PFU C15 with high encapsulation yield and optimal size for tetracy-cline (279.9 μg/mL, 13.1 nm) and cefoxitin (247.5 μg/mL, 14.8 nm), but with occurring ferritin fractions (200 – 500 nm). Then, the encapsulated antibiotics were tested on dif-ferent bacterial planktonic cells and on S. aureus biofilm. The encapsulated cefoxitin did not inhibit bacterial growth, presumably due to degradation during encapsulation pro-cess; and encapsulated vancomycin did not reduce vancomycin resistant bacteria, the nanocarrier was probably not able to overcome bacterial defense mechanisms. Encapsu-lated tetracycline had the same effect as tetracycline alone on planktonic cells and cells in S. aureus biofilm.
