Identification of bacteria associated with immune cell profile from peripheral blood

Abstract:

Colorectal cancer is still a very serious and frequent disease affecting a significant part of the Czech Republic. There is now increasing evidence on the influence of the intestinal microbiome on the processes of colorectal carcinogenesis, while it is clear that disruption of the composition of the intestinal microbiota promotes tumor progression, mainly through its interaction with components of the innate and adaptive immune system. This master thesis focuses on the identification of associations between bacteria from tumor swabs and stool samples of patients with colorectal cancer and specific populations of immune cells circulating in the peripheral blood. The first chapter of this thesis focuses on describing the theoretical background of colorectal cancer, the gut microbiota, the immune system, and the mechanisms by which the microbiome influences immune system responses. The second chapter of the thesis consists of a description of the methods used for the processing and statistical analysis of the received data. The results of the thesis show significant associations between the microbiota of tumour and stool swab samples and specific immune markers such as regulatory T-lymphocytes and NK cells. Moreover, the results of the analysis identify a potentially significant association between immune system components and the presence of oral pathogens in the gut, suggesting that the microorganisms in question might possess properties that promote tumor initiation and progression.

Abstract:

Kolorektální karcinom je nadále velmi závažným a častým onemocněním, postihujícím podstatnou část České republiky. V současnosti narůstá množství důkazů o vlivu střevního mikrobiomu na procesy kolorektální karcinognézy, přičemž je zřejmé, že narušení složení střevní mikrobioty podporuje nádorovou progresi, a to zejména prostřednictvím její interakce se složkami vrozeného a adaptivního imunitního systému. Diplomová práce se zaměřuje na identifikaci asociací mezi bakteriemi ze vzorků stěrů nádoru a stolice pacientů s kolorektálním karcinomem a specifickými populacemi imunitních buněk cirkulujících v periferní krvi. Práce je v první kapitole zaměřena na popis teoretického pozadí kolorektálního karcinomu, střevní mikrobioty, imunitního systému a mechanismů vlivu mikrobiomu na reakce imunitního systému. Druhou kapitolu diplomové práce tvoří popis metod využitých ke zpracování a statistické analýze obdržených dat. Výsledky práce poukazují na významné asociace mezi mikrobiotou vzorků stěrů nádoru a stolice a specifickými imunitními markery, jako jsou regulační T-lymfocyty a NK buňky. Na základě výsledků analýzy je navíc identifikována potenciálně významná asociace mezi složkami imunitního systému a přítomností orálních patogenů ve střevech, přičemž se domníváme, že by mohly zmiňované mikroorganismy disponovat právě vlastnostmi podporujícími nádorovou iniciaci a progresi.
