The lec1-2 Mutant: A Genetic Kaleidoscope Unleashing Unforeseen Phenotypic Abnormalities

Abstract:

T-DNA inzerční mutanti v Arabidopsis se během posledních tří dekád ukázali jako účinný nástroj umožňující významný pokrok v pochopení molekulárních mechanismů komplexních vývojových procesů ve vyšších rostlinách, jako je například vývoj květů nebo vrozená imunita. Tito mutanti avšak mají i stinnou stránku, pramenící z možnosti negativního ovlivnění cílového genomu inzercí T-DNA. Naše studie se zaměřila na zkoumání T-DNA inzerčního mutanta lec1-2. Specifický embryonální fenotyp byl charakterizován v tomto mutantu a je také spojován s mutantem lec1 (Lotan a kol., 1998; Meinke a kol., 1994). Náš výzkum ale ukázal, že oproti mutantu lec1 se lec1-2 vykazuje jedinečnou kombinací defektů květenství a transkripčního přeprogramování, které indikuje přítomnost autoimunity. Nicméně jsme ale zjistili, že alela lec1-2 není u tohoto mutanta původcem pozorovaných fenotypových změn. Celogenomové sekvenování navíc odhalilo přítomnost dalších mutací v lec1-2, které zahrnují širokou škálu variací od jednonukleotidových polymorfismů až po změny struktury genomu. Tyto genetické aberace byly nalezeny nejen v genu LEC1 ale i v genech hrajících roli v procesech jako je imunita, oprava DNA a remodelace buněčné stěny. Tyto popsané aberace v genech souvisejících s imunitou, včetně imunitních receptorů, nabízí věrohodné vysvětlení pro konstitutivní aktivaci imunitních drah. Popsané mutace genů zodpovědných za remodelace buněčné stěny a kódujících transkripční faktory mohou poskytnout molekulární základy pro pozorované fenotypy květů. Nicméně další zkoumání je zásadní pro doložení těchto zjištění a komplexní pochopení základních mechanismů.

Abstract:

Over the past three decades, Arabidopsis T-DNA insertional mutants have emerged as a potent tool, enabling significant advancements in understanding the molecular intricacies of complex developmental processes in higher plants, such as flower development and innate immunity. However, these mutants do come with a significant drawback stemming from the potential damage caused by T-DNA insertions in the plant genome. Our study focused on investigating a specific T-DNA insertional mutant, the leafy cotyledon 1-2 (lec1-2) mutant. The lec1-2 mutant has long been recognized for its characteristic embryonic phenotypes associated with lec1 mutants (Lotan et al., 1998; Meinke et al., 1994). However, our research revealed that the mutant exhibits a unique combination of flower defects and transcriptional reprogramming indicative of autoimmunity, distinguishing it from the typical lec1 mutants. We established that the observed phenotypic alterations in the mutant are not attributable to the lec1-2 allele. Furthermore, whole genome sequencing analysis identified numerous other mutations within the mutant, encompassing a wide range from single nucleotide variations to structural variations. These genetic variations extend beyond the LEC1 gene and encompass genes involved in processes such as immunity, DNA repair, and cell wall remodeling. The presence of genetic variations in defense-related genes, including immunity receptors, offers a plausible explanation for the constitutive activation of immunity pathways. Additionally, mutations in multiple genes including cell wall remodelling genes and transcription factors may provide molecular basis for the observed flower phenotypes. Nevertheless, further investigation is crucial to substantiate these findings and comprehensively understand the underlying mechanisms.
