The occurence of Antibiotic resistance genes (ARGs) in two stages of wastewater treatment in a local Wastewater treatment plant (WWTP)

Abstract:

Tato práce se zabývá rozšířením rezistence k antibiotikům a genů, jež tuto rezistenci kódují (ARGs), v čistírnách odpadních vod (ČOV), jež jsou považovány za významný zdroj tohoto znečištění v životním prostředí. Pozornost je věnována zejména ?-laktamázovým genům. Teoretická část je zaměřena na jednotlivé mechanismy antibiotické rezistence u bakterií. Shrnuje rozšíření ARGs a bakterií rezistentních k antibiotikům (ARBs) v různých prostředích, jako jsou povrchové vody, půda a zejména čistírny odpadních vod (ČOV). Mezi nejčastěji studované a v literatuře popisované geny patří právě geny kódující ?-laktamázy, které představují vážné epidemiologické riziko v humánní medicíně. Popisovány jsou také faktory, které ovlivňují osud ARGs a ARBs v bakteriálním rezistomu jednotlivých prostředí. V problematice ČOV je diskutován především vliv struktury molekuly antibiotika a typu ČOV na efektivitu přečištění odpadních vod od ARGs. Experimentální část práce popisuje konkrétní situaci v bakteriální populaci ČOV v Ostravě z hlediska přítomnosti ARGs v aktivační a dosazovací nádrži. Geny blaTEM a tetW byly detekovány v obou nádržích po celou dobu vzorkovací kampaně. U ampicilin rezistentních izolátů z posledního odběru byla stanovoven hodnota IC50 pro ampicilin. Izoláty z dosazovací nádrže vykazovaly vyšší hodnoty IC50 (33 % izolátů vykazovalo hodnoty IC50 > 10 mg/ml) než izoláty z aktivační nádrže (92 % izolátů vykázalo hodnoty IC50 < 4 mg/ml). Tento výsledek ukazuje na význam procesů v dosazovací nádrži v selekci ampicilin rezistentních kmenů bakterií.

Abstract:

This work is focused on the dissemination of bacterial resistence to antibiotics and genes encoding the antibiotic resistence (ARGs) in the wastewater treatment plants (WWTPs), which are considered as a main source of ARGs in the environment. Special attention is paid to the genes encoding bacterial ?-lactamases that pose a serious epidemiological risk for human medicine. The theoretical part of the work summarizes the individual mechanisms of bacterial resistence towards antibiotics and the spread of ARGs and resistant bacteria (ARBs) in environmental compartments, like river waters, soil, and WWTPS. It was repetadelly described in the literature that the genes encoding ?-lactamases belonged to the most frequently described ARGs in the environment. The factors affecting the fate of ARGs and ARBs in the environment are also discussed in this part of the work. In case of WWTPs, those are primarily the chemical structure of the antibiotic molecule and WWTP processes involved in the wastewater treatment. They affect the ARGs removal effectivity of WWTPs. The experimental part of this work studies the bacterial resistome of a WWTP located in Ostrava, Czech Republic. The genes blaTEM and tetW were detected in the bacterial population of its nitrification tank and that of the sedimentation tank during the 1-year time of sampling. Ampicillin resistant isolates were tested for their IC50 values for Ampicillin. The bacterial strains isolated from the sedimentation tank showed higher IC50 values (33 % of isolates showed IC50 values > 10 mg/ml) compared to those obtained from the nitrification tank (92 % of isolates showed IC50 values < 4 mg/ml). It indicates the processes in the secondary sedimentation phase of the wastewater treatment play an important role in selection of ampicilin resistant bacteria.
