Bone Development Signaling Pathways and Their Relevance for Progression of Bone Malignancies

Abstract:

Proteiny a jimi regulované signální dráhy jsou zapojeny nejen do fyziologických, ale i patologických biologických procesů. Tato disertační práce se věnuje úloze vybraných proteinů v regulaci rozvoje osteosarkomu i fyziologické homeostáze kostní tkáně. V první části identifikuje protein c-Myb jako negativní prognostický marker pro pacienty s osteosarkomem a experimentálně potvrzuje jeho onkogenní a prometastatický charakter při rozvoji tohoto onemocnění. Následně se zabývá nalezením dalších markerů progrese osteosarkomu. Ukazuje protein PYCARD jako možný nádorový supresor a pozitivní prognostický marker a popisuje jeho význam v regulaci citlivosti osteosarkomových buněk k chemoterapeutikům. Druhá část práce pak identifikuje kaspázu 9 jako protein regulující migraci osteoblastů a popisuje potenciální nový substrát tohoto enzymu, protein ABHD2. Tato práce tedy předkládá nové poznatky o osteosarkomu na molekulární úrovni s možným přesahem do diagnostické a terapeutické onkologické péče a přispívá k detailnějšímu objasnění molekulárních mechanismů účastnících se regulace fyziologické migrace osteoblastů.

Abstract:

Proteins and signaling pathways are involved not only in physiological but also in pathological biological processes. This thesis describes the role of selected proteins in the regulation of osteosarcoma progression and physiological bone homeostasis. Firstly, it defines protein c-Myb as a negative prognostic marker in osteosarcoma and confirms its oncogenic and pro-metastatic role using various experimental approaches. Subsequently, it identifies other markers of osteosarcoma progression and shows protein PYCARD as a possible tumor suppressor and positive prognostic marker that is involved in the regulation of chemosensitivity of osteosarcoma cells. The second part of this thesis describes protein caspase 9 as a regulator of osteoblastic cell migration and identifies protein ABHD2 as a possible substrate of this protease. This thesis presents new findings about osteosarcoma at the molecular level with possible implications in the diagnostic and therapeutic oncological care and contributes to the elucidation of molecular mechanisms involved in the regulation of physiological migration of osteoblasts.
