Methods for targeted genetic modifications of microalgae for improvement of their biotechnological potential

Abstract:

Tato rešeršní bakalářská práce se zabývá shrnutím metod cílených genetických modifikací mikrořas a jejich potenciálem pro inovaci biotechnologických aplikací. Práce poskytuje přehled o biotechnologickém využití mikrořas ve farmaceutickém průmyslu, potravinářství a energetice. Rovněž představuje metodické přístupy ke genetickým úpravám těchto organismů. Klade důraz na mechanismy přípravy DNA konstruktů pro vložení do genomu mikrořas a jejich strukturu, techniky doručení cizorodé DNA do buněk, cílené zásahy do genomu a metody pro selekci modifikovaných buněk. V práci jsou detailně prozkoumány současné přístupy a technologie v genetickém inženýrství, zejména využití nástrojů jako CRISPR/Cas9, které umožňují přesné zásahy do genomu mikrořas. Cílem této bakalářské práce je shrnout a analyzovat dostupné technologie a metodiky, které mohou přispět k efektivnějšímu a širšímu využití mikrořas v biotechnologickém průmyslu.

Abstract:

This research bachelor thesis summarizes methods for targeted genetic modifications of microalgae and their potential for innovations in biotechnological applications. The thesis provides an overview of the biotechnological applications of microalgae in the pharmaceutical, food and energy industries. It also presents methodological approaches to genetic modification of these organisms. It emphasizes the mechanisms of preparation of DNA constructs for insertion into the microalgal genome and their structure, techniques for delivery of foreign DNA into cells, targeted genome interventions and methods for selection of modified cells. Current approaches and technologies in genetic engineering are explored in detail, particularly the use of tools such as CRISPR/Cas9, which enable precise interventions in the microalgal genome. The aim of this bachelor thesis is to summarise and analyse the available technologies and methodologies that can contribute to a more efficient and wider use of microalgae in the biotechnology industry.
