Effects of temperature and light on xanthophyll cycle activity. (Analysis of response variability among selected plant species.)

Abstract:

Diplomová práce je zaměřena na analýzu vlivu fotosynteticky aktivní radiace a zvýšené teploty na dynamiku deepoxidační reakce xantofylového cyklu. Hlavním cílem bylo posoudit rozdíly v odezvě xantofylového cyklu na teplotu u různých druhů rostlin. Pro účely experimentu byly vybrány semenáčky smrku ztepilého (Picea abies L. Karsten) a rostliny Arabidopsis thaliana WT. Rostliny byly pěstovány v růstových komorách při ozářenosti FAR 50 ?mol.m-2.s-1. Měření byla prováděna na temnostně adaptovaných jehlicích či listech, které byly po dobu 5 minut aklimovány na požadovanou teplotu z intervalu 20 °C až 40 °C. Pro posouzení vlivu světla byla vybrána saturační ozářenost 1400 ?mol.m-2.s-1 a délky ozáření v intervalu od 1 s až po několik desítek minut. Zastoupení fotosyntetických pigmentů ve vzorcích bylo stanovováno HPLC analýzou. Vliv světla a teploty byl posuzován na základě deepoxidačního stavu pigmentů xantofylového cyklu (DEPS, %). Na základě provedených experimentů jsme se pokusili ověřit hypotézu, že zvýšená teplota urychlí světlem indukovanou deepoxidaci více u jehlic smrku než u listů A. thaliana. U smrku došlo k urychlení deepoxidační reakce a zvýšení DEPS již při teplotě 32 °C a délce ozáření 10 s. Ještě výraznější urychlení bylo pozorováno při deseti po sobě jdoucích 1 s světelných pulzech v průběhu 1 minutového intervalu (10x1s). Zvýšení hodnot DEPS bylo také doprovázeno výraznou akumulací Zx, zejména v případě deseti 1 s pulzů. Pouze v případě smrku bylo také pozorováno výrazné zvýšení DEPS při teplotě 40 °C již po jednom 1 s světelném pulzu. U A. thaliana bylo urychlení deepoxidační reakce pozorováno až při teplotě 40 °C. Navíc při 10 s ozařování (nebo deseti 1 s pulzech) nebyla pozorována přítomnost Zx, což naznačuje, že v listech A. thaliana byl urychlen pouze první krok deepoxidační reakce. Prokázali jsme tedy, že zatímco u A. thaliana dochází k výraznému urychlení prvního kroku deepoxidační reakce skokem při 40 °C, u jehlic smrku ztepilého je velmi rychlá konverze violaxantinu na zeaxantin stimulována postupně s rostoucí teplotou. Předpokládáme, že k usnadněné deepoxidaci violaxantinu při zvýšené teplotě přispívá rozdílná teplotní závislost fluidity tylakoidních membrán jehlic smrku ve srovnání s listy A. thaliana.

Abstract:

Presented thesis is focused on the analysis of the impact of photosynthetically active radiation and elevated temperatures on the dynamics of violaxanthin deepoxidation. The aim was to evaluate the differences of temperature response of xanthophyll cycle in the various plant species. For the experiment seedlings of Norway spruce (Picea abies L. Karsten) and wild type of Arabidopsis thaliana have been selected. Plants were grown at 50 ?mol.m-2.s-1 irradiance of PAR in growth chamber. Dark-adapted samples (needles or leaves) preaclimated to the desired temperature in the range 20 °C - 40 °C for 5 minutes were used for the measurements. In order to assess the impact of irradiance saturation light intensity 1400 ?mol.m-2.s-1 and the irradiation regimes in the range from 1 second up to several tens of minutes were applied. Content of photosynthetic pigments was determined by HPLC analysis. The impact of light and temperature was evaluated through deepoxidation state of xanthophyll cycle pigments (DEPS, %). Based on the above mentioned experiments the hypothesis was tested that elevated temperatures accelerate light induced violaxanthin deepoxidation to a greater extent in Norway spruce needles than in A. thaliana leaves. In spruce needles violaxanthin deepoxidation and DEPS was stimulated already at the temperature 32 °C and 10 seconds of irradiation. Even more pronounced stimulation of deepoxidation was observed after 10 light-pulses of 1s duration in 1 min interval (10x1s). Enhanced DEPS was also accompanied by accumulation of considerable amount of Zx, particularly in 10x1s regime of irradiation. Only in spruce a significant increase of DEPS was observed already after one 1 s light pulse at 40 °C. In A. thaliana leaves significant acceleration of V deepoxidation occurred just at 40°C. However, no presence of Zx was observed after 10 s irradiation (or 10 light-pulses of 1s duration) at 40 °C, suggesting that only the first step of Vx deepoxidation was accelerated in A. thaliana leaves. We have proved that, whereas in A. thaliana the first step of violaxanthin deepoxidation is significantly accelerated step-wisely at 40 °C, in spruce needles extremely rapid conversion of violaxanthin to zeaxanthin is stimulated gradually upon increasing temperature. We suggest that different temperature dependence of thylakoid membrane fluidity between spruce needles and A. thaliana leaves contributes to facilitated violaxanthin deepoxidation at elevated temperatures in spruce needles.
