Diplomová práce se zabývá spektrofotometrickým a hmotnostně-spektrometrickým studiem reakce derivatizace oligonukleotidů obsahujících thymin (AAAAT, AATAA, AAAATT, AATTAA, ATATAA, ATAATA) komplexem OsO4-bipyridin (OsBipy). Spektrofotometricky bylo potvrzeno, že se tvoří více produktů a pomocí MALDI TOF MS bylo zjištěno, že OsBipy reaguje především s thyminovou bází a oxiduje ji za vzniku di-hydroxy …more
Abstract:
Thesis deals with spectrophotometric and mass-spectrometric study of the derivatization reaction of thymine containing oligonucleotides (AAAAT, AATAA, AAAATT, AATTAA, ATATAA, ATAATA) by OsO4-bipyridine complex (OsBipy). By spectrophotometry it was proved that several products are formed and with the help of MALDI TOF MS it was found that OsBipy reacts first of all with thymine base and oxidizes it …more
