Heteroborane derivatization of alcohols

Abstract:

Tato bakalářská práce se zabývá syntézou šesti referenčních analytických standardů derivátů unikátně reaktivního heteroboranu [8-(1,4-dioxan-1-ium)-kloso-1,2-dikarba-3-kobaltadodekaboran(10)-3-komo-3'-[kloso-1',2'-dikarba-3'-kobaltadodekaborát(11)] (C8D), který je testován jako nové, bezpečnější činidlo pro předkolonovou HPLC derivatizaci alkoholů, zejména pro případ, že není k dispozici GC nebo je alkohol málo těkavý nebo tepelně nestálý. Reakcí C8D v nadbytku alkoholu v roli rozpouštědla a reaktantu zároveň s kovovým sodíkem v inertní atmosféře za standardních podmínek se podařilo získat pět ze šesti derivátů alkoholů ve vynikajících výtěžcích. Dle očekávání nejrychleji v řádu minut reagoval methanol, nejpomaleji v řádu desítek hodin glycerol. Struktura všech připravených derivátů cesných solí byla potvrzena 11B, 13C, 1H NMR a MS. Čistota produktů byla ověřena s pomocí HPLC PDA MS, stanovením bodu tání, hmotnostní spektrometrií a elementární analýzou CHN. Pouze monohydroxyalkoholy a ethylenglykol reagovaly při laboratorní teplotě s C8D za vzniku derivátů, které byly detekovány za pomoci HPLC PDA MS.

Abstract:

This bachelor's thesis deals with the synthesis of six reference analytical standards of derivatives of the uniquely reactive heteroborane [8-(1,4-dioxan-1-ium)+-closo-1,2-dicarba-3-cobaltadodecaborane(10)-3-commo-3'-[closo-1',2'-dicarba-3'-cobaltadodecaborate(11)-] (C8D), which is being tested as a new, safer reagent for pre-column HPLC derivatization of alcohols, especially when GC is not available or alcohol is low volatility or thermally unstable. C8D reacts in an excess of alcohol used simultaneously as a solvent and reactant with metallic sodium in an inert atmosphere under standard conditions. Five from six studied alcohols reacted to obtain isolated their C8D derivatives in excellent yields. As expected, methanol reacted in the order of minutes, glycerol slowly in the order of tens of hours at standard conditions. The structure of all prepared cesium salt derivatives was confirmed by 11B, 13C, 1H NMR and MS. The purity of the products was verified with the help of HPLC PDA MS, determination of the melting point, mass spectrometry and elemental analysis of CHN. A model mixture of alcohols with water was mixed together with C8D in order to test the use of C8D as precolumn derivatization reagent for the qualitative analysis of alcohols in aqueous solution. Monohydroxy alcohols and diethylenglycol only were detected after the reaction with room temperature reaction with C8D and the derivatives were successfully detected by the HPLC PDA MS.
