Interaction of mammalian oocytes and sperm in relation to in vitro fertilization

Abstract:

Cílem disertační práce bylo studovat expresi glykoproteinů zona pellucida (ZP2, ZP3) během procesu zrání a primárního kontaktu bovinních oocytů a kapacitovaných spermií. Charakterizovat interakci gamet v modifikovaném kokultivačním prostředí z aspektu oplození v systému in vitro. Oocyty s vyšší kompetencí exprimovaly ZP2 protein jak ve stadiu GV, tak i MII, méně kompetentní oocyty syntetizovaly ZP2 protein pouze ve stadiu GV. Meioticky méně kompetentní oocyty exprimovaly ZP3 protein ve stadiu GV i MII, protein ZP3 však nebyl nalezen po kokultivaci zralých oocytů a akrozom reagujících spermií. Kinetika a účinnost oplození i vývoj embryí byly odlišné, pokud byly oocyty kokultivovány se spermiemi v modifikovaných systémech. Kokultivační prostředí, ve kterém dochází k interakci obou gamet, významně ovlivňuje proces oplození in vitro a vývoj raných bovinních embryí.

Abstract:

The aim of this dissertation was to study the expression of zona pellucida glycoproteins (ZP2, ZP3) during the maturation process and the primary contact of bovine oocytes and capacitated sperm. To characterize the interaction of gametes in a modified co-culture environment from the perspective of fertilization in an in vitro system. The more competent oocytes expressed ZP2 protein at both GV and MII stages, the less competent oocytes only synthesized ZP2 protein at the GV stage. Meiotically less competent oocytes expressed ZP3 protein at both GV and MII stages, however ZP3 protein was not detected after the co-culture of matured oocytes and acrosome reacted sperm. Fertilization kinetics, efficiency and embryo development differed when oocytes were co-cultured with sperm in modified systems. The co-culture environment, in which both gametes interact, significantly influences the process of in vitro fertilization and early bovine embryo development.
