Abstract:

Srovnávací morfologie vývojových stadií gregarin a kryptosporidií se zaměřením na interakce mezi hostiteli a parazity Andrea Valigurová Gregariny a kryptosporidie jsou parazitická protista kmene Apicomplexa, která upoutala velkou pozornost především po provedených fylogenetických analýzách, které prokázaly jejich blízkou příbuznost. Gregariny i kryptosporidie jsou díky svému unikátnímu způsobu přichycení k hostitelské buňce a následnému vývoji považovány za zvláštní skupinu parazitických protist. Interakce mezi hostitelem a parazitem a strategie parazita při přichycení k hostitelské buňce můžeme zařadit mezi významné charakteristiky při posuzování fylogenetických vztahů mezi gregarinami a kryptosporidiemi. Předkládaná práce se zabývá posouzením počátečního vývoje sledovaných parazitů v hostiteli na ultrastrukturální úrovni a hledáním společných znaků pro oba taxony protistů. V průběhu této studie byly provedeny experimentální infekce hostitelů s cílem charakterizovat kontrolovaný parazito-hostitelský model, který bude vhodný pro studium počátečního vývoje gregarin a kryptosporidií. Výsledkem této práce jsou publikované morfologické studie čtyř druhů eugregarin, Gregarina steini, Gregarina polymorpha, Gregarina garnhami a Leidyana ephestiae, které parazitují v laboratorních chovech hmyzu. Pozornost byla zaměřena především na ultrastrukturu a postupný vývoj epimeritu, jakož i jeho interakce s hostitelským epitelem. Shodné výsledky byly získány studiem eugregarin Gregarina cuneata a Gregarina niphandrodes (nepublikovaná data). U kryptosporidií byl sledován proces uchycení a počáteční vývoj v hostiteli u dvou evolučně vzdálených druhů žaludečních kryptosporidií: Cryptosporidium muris u experimentálně infikovaných hlodavců a Cryptosporidium sp. ‘toad’ u žáby. U obou druhů byly pozorovány změny mikroklků hostitelské buňky a následné vytvoření záhybu cytoplasmatické membrány, který postupně obrůstá parazita a vytváří parazitoforní vak. Na základě těchto výsledků navrhujeme použití termínu „epicelulární“ pro vhodnější vyjádření lokalizace kryptosporidií v hostiteli, místo doposud užívaného termínu „intracelulární extracytoplasmatická“. Pro potvrzení této hypotézy byly studovány kontrolní organizmy, střevní kryptosporidie Cryptosporidium parvum z hlodavců, Cryptosporidium saurophilum a Cryptosporidium sp. z ještěrů, a bylo dosaženo shodných výsledků. U gregarin i kryptosporidií se apikální komplex ztrácí již v počátcích interakce s hostitelskou buňkou a oba paraziti se přichycují k hostitelské buňce pomocí apikálních výběžků, epimeritem u eugregarin a přichycovací organelou s přídavnými strukturami u kryptosporidií. V práci jsou detailně popsány morfologické změny, ke kterým dochází při vzniku epimeritu, které jsou porovnány s procesem tvorby přichycovací organely u kryptosporidií. Strategie přichycení obou sledovaných protistů k hostitelské buňce je velmi podobná. Oba organizmy mají epicelulární lokalizaci mezi mikroklky epiteliálních buněk hostitele, přičemž eugregariny nejsou obaleny záhybem cytoplasmatické membrány hostitelské buňky. U vývojových stádií neogregregariny Mattesia dispora byla studována ultrastruktura a zvláštní pozornost byla věnována vzniku oocyst. Podobně jako eugregariny je i tato neogregarina velmi podobná kryptosporidiím, především schopností autoinfekce oocystami a chováním vývojových stadií během životního cyklu. Bez ohledu na některé zjištěné rozdíly mezi gregarinami a kryptosporidiemi, výsledky této studie podporují fylogenetickou příbuznost obou sledovaných skupin protistů.

Comparative morphology of developmental stages of gregarines and cryptosporidia with an emphasis on host-parasite interactions Andrea Valigurová Gregarines and cryptosporidia are parasitic protists belonging to the phylum Apicomplexa. They have recently gained much attention since some phylogenetic analyses showed that cryptosporidia are more closely related to the gregarines than to coccidia. Both of these apicomplexans are often considered to be peculiar organisms, as they exhibit some unique features in their attachment strategy and development. In addition, cryptosporidia are similar to gregarines in many developmental aspects. As the host-parasite interactions and attachment strategy both seem to be significant markers for tracing the phylogenetic relationships among gregarines and cryptosporidia, this study focuses mainly on the ultrastructural analysis of their early development and searches for explicit characteristics common to both apicomplexans. In order to obtain a controllable host-parasite system, suitable for studies on early development of gregarines and cryptosporidia, experimental infections were carried out. Morphological data on four eugregarine species, Gregarina garnhami, Gregarina polymorpha, Gregarina steini, and Leidyana ephestiae, parasitic in laboratory reared insects, are presented in the publications listed in thesis. Particular attention was given to the gradual development and fine structure of the epimerite and its relationship to the host epithelium. Additional unpublished data on Gregarina cuneata and Gregarina niphandrodes generally support observations made on the already mentioned eugregarine species. As for cryptosporidia, the invasion process and early development were observed at the ultrastructural level in two evolutionarily distinct gastric species, Cryptosporidium muris from experimentally infected rodents and Cryptosporidium sp. ‘toad’ from naturally infected toads. In both cryptosporidia, the contact between the invading zoite and the host cell induces recruitment of the host microvillar membrane, which loses its microvillous appearance and forms a tight-fitting elastic membrane fold gradually rising along the parasite, eventually forming the parasitophorous sac. Data presented herein support the term epicellular to reflect cryptosporidian localization, rather than the term intracellular-extracytoplasmic traditionally used throughout the literature. Additional observations on intestinal cryptosporidia (Cryptosporidium parvum from rodents, Cryptosporidium saurophilum and Cryptosporidium sp. from lizards) were used as controls in order to support the hypothesis of this work. In both eugregarines and cryptosporidia, the apical complex disappears early during cell invasion and they are attached to the host cell by their apical processes, i.e. epimerite in eugregarines and the feeder organelle with associated structures in cryptosporidia. The basic changes that the epimerite undergoes during its formation are described and compared with the attachment process and feeder organelle formation in cryptosporidia. The epimerite region and whole attachment site of eugregarines resemble the feeder organelle with associated structures in cryptosporidia. The cryptosporidian attachment strategy is very similar to that of the eugregarines, which are also epicellularly located on the microvillous surface of host epithelial cells, although not enveloped by host cell membrane folds. In addition to studies on eugregarines, the developmental stages of the neogregarine, Mattesia dispora, have been studied, focusing mainly on oocyst development. Similar to eugregarines, this neogregarine also exhibits some striking similarities with cryptosporidia, especially in the presence of autoinfection and the behaviour of their developmental stages during the life cycle.
