Tato práce popisuje metodiky vhodné k izolaci chloroplastů a thylakoidních membrán z intaktních jehlic smrku ztepilého [Picea abies (L.) Karst]. Hlavním cílem je vytvořit metodiku, která povede k izolaci thylakoidních membrán, jejichž fluorescenční parametr FV/FM se bude co nejvíce přibližovat hodnotě parametru FV/FM na původních intaktních jehlicích. Abstract:
This thesis describes procedures for isolation of chloroplasts and thylakoid membranes from intact Norway spruce needles [Picea abies (L.) Karst]. The main objective of this thesis is suggest the method, which leads to isolation of thylakoid membranes with the highest value of the fluorescent parameter FV/FM, which can be same as the value of the parameter FV/FM of the intact needles.
