Molecular mechanisms controlling chromosome segregation in ooocytes and early embryos

Abstract:

Je tomu již více než osmdesát let od doby, kdy bylo poprvé popsáno spojení mezi chromozomálními abnormalitami a klinickým onemocněním u Downova syndromu. Od té doby byly aneuploidie popsány jako hlavní příčina potratů, abnormálního průběhu těhotenství a vrozených vývojových vad u lidí. Původní studie prokázaly, že většina aneuploidií pochází ze samičí meiózy a že jejich frekvence stoupá se zvyšujícím se věkem matky. Meióza je základním procesem, kterým musí projít gamety u obou pohlaví. V principu je schopnost těchto specializovaných buněk redukovat počet svých chromozomů na polovinu závislá na správném párování homologních chromozomů. Jednou z nejdůležitějších struktur, podílejících se na správném rozdělení chromozomů, je dělicí vřeténko. V mitotických buňkách a v oocytech nižších organizmů (např. u mořských hvězdic a ježovek) slouží jako nukleační centra a základní struktury pólů dělicího vřeténka centrozomy. Nicméně u strunatců, hmyzu a C. elegans jsou centrozomy eliminovány v časné fázi vývoje oocytu a dělicí vřeténko se u nich sestavuje acentrozomálním způsobem. Konkrétně u myší vzniká vřeténko v meióze a během prvních několika embryonálních dělení fúzí mnoha nukleačních center. Předkládaná doktorská práce se zabývá čtyřmi rozdílnými, ale úzce příbuznými tématy. V první části je popsán vliv kultivace in vitro na morfologické parametry meiotického dělicího vřeténka u myších oocytů. V našich experimentech jsme prokázali výraznou akumulaci molekulárního motorového proteinu kinezinu 5 (Eg5, Kif11) v oblasti pólů dělicího vřeténka u in vitro kultivovaných buněk. Tyto byly taktéž více citlivé k působení inhibitoru Eg5, což naznačuje možný mechanizmus udržování stability dělicího vřeténka, závislý na tomto proteinu. Druhá část je zaměřena na vliv genetického pozadí na přirozenou úroveň aneuploidie u in vitro maturovaných oocytů vybraných kmenů myší. V této studii byla použita technika počítání kinetochor v intaktních buňkách s cílem změřit míru aneuploidie pocházející z prvního meiotického dělení. K tomuto účelu byly vybrány tři kmeny – CD-1, C57/Bl6 a C3H/NeJ. Výsledky jasně prokázaly predispozici určitých kmenů k předčasnému rozestupu sesterských chromatid. Třetí kapitola popisuje sterilitu mezidruhových hybridů a roli samičí meiózy v tomto fenoménu. Prezentované výsledky jsou zaměřeny na defekty kongrese v prvním meiotickém dělení a na její přímé důsledky – přítomnost celých nespojených bivalentů ve druhém meiotickém dělění a prodloužené vydělování prvního pólového tělíska. V poslední části jsou prezentována data týkající se otázky regulace délky dělicího vřeténka u dvoubuněčných myších embryí. Během prvních několika embryonálních dělení se buňky dělí, aniž by následně zvětšovaly objem. Tato změna ve velikosti se musí projevit ve změně délky dělicího vřeténka. U Xenopus laevis a myšího embrya byl navržen model, kdy dělicí vřeténko dosahuje horního limitu své délky na začátku vývoje. Na základě našich dat je spíše pravděpodobné, že dvoubuněčné myší embryo přesně kontroluje délku svého dělicího vřeténka na základě poměru mezi objemem jádra a objemem cytoplazmy

Abstract:

The link between the clinical disorder named Down’s syndrome and the chromosome abnormality has been described for the first time some 80 years ago. Since that time, it has been established that it is the aneuploidy which is the leading cause of pregnancy loss, abnormal pregnancy and congenital birth defects in humans. Even the results of early studies revealed that the frequency of aneuploidy is higher during the oogenesis in comparison to spermatogenesis, and it exhibits positive correlation with the maternal age. Meiosis is a special cell division type which occurs during both – oogenesis and spermatogenesis. For both types of the gametes, in order to reduce the number of the chromosomes to a half of the original amount, formation of a correct pairwise association between homologous chromosomes is required. Oocytes and sperm therefore exhibit unique structural and functional properties, which are essential for the coordination of the complex processes of meiosis. One of the most important “tools” for proper chromosome distribution during cell division is the spindle apparatus. It is essential for chromosome attachment, alignment and segregation. The centrosomes, which are nucleation centers of microtubules and basic pole structures, are present in mitotically dividing cells and oocytes of lower organisms, such as sea urchins and starfish. However, in C. elegans, insects and chordates, centrosomes are eliminated during oogenesis before the meiotic division and the spindle is established in an acentrosomal manner. In mice, the meiotic spindle and the spindles in several first mitotic divisions of the early embryonic stage are formed through the fusion of multiple microtubules organizing centers. The presented PhD thesis focuses on four different, but closely related questions. The first part of the thesis is dedicated to the effect of in vitro culture on morphological parameters of the meiotic spindle in mouse oocytes. Our experiments revealed that Eg5 (Kif11) molecular motor is accumulated on the spindle poles more densely in in vitro matured MII oocytes in comparison to in vivo matured oocytes. These cells were also more sensitive to the Eg5 inhibitor compared to the in vivo control group, which indicates the Eg5-dependent mechanism of the spindle maintenance. In the second part, we describe the influence of genetic background on aneuploidy incidence in selected commonly used mouse strains. We established the levels of aneuploidy, resulting from the missegregations in meiosis I in three mice strains – CD-1, C57BL6, and C3H/HeJ. The results demonstrate the predisposition of certain strains to the premature sister chromatid segregation. For the purposes of this study, we have used a modified, highly precise kinetochore counting technique. The third part is focused on interspecific hybrid sterility and the contribution of female meiosis to this phenomenon. The presented data reveal congressional defects in meiosis I in oocytes of hybrid females as well as the direct consequences of these defects – whole bivalent nondisjunction in meiosis II and a delayed first polar body extrusion in hybrid group in comparison to controls. The last presented paper addresses the question of spindle size regulation in 2-cell mouse embryos. It is known that cells in early embryos decrease in volume. This change in size of the cells must be reflected in the spindle scaling. It has been suggested that in Xenopus and mouse embryos, spindle length reaches its upper limit in large cells at the beginning of the development. Our experiments with 2-cell mouse embryos have shown that the spindle length is conditioned by the ratio between the nuclear and the cytoplasmic volumes.
