Comparison of Three Methods of Anaerobic Pathogenic Bacteria Cultivation

Abstract:

Nezbytným předpokladem pro úspěšný průkaz anaerobních patogenních mikroorganismů ze vzorků klinického materiálu je způsob jeho kultivace. V současné době se k těmto účelům používá několik způsobů kultivace v anaerobním prostředí. Jedná se o kultivaci v anaerobním boxu, v anaerobních hrncích a v jednorázových kultivačních soupravách. Cílem bakalářské práce bylo porovnat tyto běžně používané způsoby kultivace prostřednictvím růstu běžně izolovaných anaerobních patogenů na rutinně používaných kultivačních médiích. Jako testovací mikroorganismy byly použity kmeny bakterií Bacteroides fragilis, Clostridium difficile a Clostridium perfringens. Jako testovací kultivační média byly použity půdy Schaedler s beraní krví a Wlikins-Chalgren s beraní krví a bez ní. Kvalita růstu jednotlivých bakterií byla hodnocena pomocí velikosti kolonií, ve kterých testované mikroorganismy vyrůstaly na jednotlivých kultivačních médiích po třech rozdílných způsobech anaerobní kultivace. Výsledky této práce svědčí o tom, že nejlepším způsobem anaerobní kultivace je kultivace v prostředí anaerobního boxu. Jako alternativní způsob kultivace lze stejně dobře použít kultivaci v jednorázových kultivačních soupravách a kultivaci v anaerobních hrncích. Kultivace v jednorázových kultivačních soupravách byla v některých případech, zvláště při kultivaci náročného anaerobního mikroorganismu Clostridium difficile lepší než kultivace v prostředí anaerobního hrnce.

Abstract:

The necessary presumption for a successful proof of anaerobic pathogenic organisms from clinical material samples is the way of its cultivation. Nowadays several cultivation ways in anaerobic atmosphere are used for these purposes. It means a cultivation in an anaerobic box, in anaerobic jars and in one-way cultivation sets. The aim of this bachelor thesis was to compare these normally used cultivation ways by means of the growth of usually isolated anaerobic pathogens on routinely used cultivation mediums. Bacterial tribes such as Bacteroides fragilis, Clostridium difficile a Clostridium perfringens were used as testing microorganisms. As testing cultivation mediums the soils Schaedler with ram blooth and Wlikins-Chalgren with ram blooth and without it were used. The growth quality of individual bacteries was estimated by the help of colonies size, in which the tested microorganisms were growing on individual cultivation mediums, always in 3 various ways of anaerobic cultivation. The results of this thesis prove that the best way of anaerobic cultivation is the cultivation in the anaerobic box atmosphere. But also the cultivation in the one-way cultivation sets and in the anaerobic pots can be used as an alternative cultivation way. The cultivation in the one-way cultivation sets, mainly during the cultivation of the difficult microorganism Clostridium difficile, was in some cases better than the cultivation in the anaerobic pot atmosphere.
