Changes in plant plasma membrane proteome and phosphoproteome induced by priming agents

Abstract:

Napadení rostliny patogenem vede ke spuštění obranných mechanizmů bránících dalšímu rozvoji infekce, kdy látky indukující obrannou reakci u rostlin nazýváme elicitory. Mezi elicitory můžeme zařadit i látky schopné aktivovat tzv. obranné nabuzení (priming) vedoucí k rychlejší a efektivnější odpovědi rostliny na stres. Při obranném nabuzení zřejmě dochází ke změně fosforylace některých proteinů a změnám proteomu plazmatické membrány rostlin. Tato diplomová práce se věnuje sledování těchto změn u buněk tabáku po aplikaci β-aminomáselné kyseliny (BABA), která indukuje u celé řady rostlin fenomén obranného nabuzení spojený s vysokou mírou rezistence proti širokému spektru patogenů. Fenomén obranného nabuzení byl studován na buňkách tabáku (Nicotiana tabacum cv. Xanthi), kdy k elicitaci byl použit proteinový elicitor kryptogein, houbový sterol ergosterol a oligogalakturonidy vznikající při degradaci rostlinných buněčných stěn. Vyvolání časné fáze obranné reakce bylo sledováno pomocí měření produkce reaktivních forem kyslíku (ROS) buňkami tabáku, na které byla 24 hodin před přidáním jednotlivých elicitorů aplikována BABA. Dále byla na buňkách sledována forsforylace Tyr zbytků kinázy SIPK pomocí metody western blot a byla měřena exprese vybraných genů zapojených v obranné reakci. Nakonec byla provedena izolace plazmatické membrány pro měření změn proteomu pomocí LC-MS/MS analýzy. Výsledky prokázaly u všech studovaných elicitorů výrazné navýšení produkce ROS po aplikaci BABA, které však zřejmě není způsobeno pre-aktivací signálních drah nebo zvýšenou expresí obranných genů v důsledku aplikace BABA. Hlavním důvodem zvýšení by tak mohlo být zvýšení koncentrace proteinů zajišťujících percepci elicitorů na plazmatické membráně. Z časových důvodů však již nebylo možné dokončit proteomickou analýzu izolovaných plazmatických membrán buněk tabáku.

Abstract:

Invasion of the plant with the pathogen results in triggering of defense mechanisms restricting further progression of the infection. Substances inducing defense reaction in plants are called elicitors. Among the elicitors are considered substances capable of activating defense priming resulting in a faster and more effective plant response to various stress conditions. The phenomenon of defense priming probably includes changes in protein's phosphorylation, as well as some changes in plasma membrane of plant cells. This thesis aims to study changes in β-aminobutyric acid (BABA) treated tobacco cells responsible for defense priming phenomenon associated with a high level of resistance to a wide range of pathogens. The defense priming phenomenon was studied on tobacco cells (Nicotiana tabacum cv. Xanthi). For elicitation protein elicitor cryptogein, fungal sterol ergosterol, and oligogalacturonides resulting from the plant cell wall degradation were used. The induction of the early phase of the defensive reaction was monitored by measuring the production of Reactive Oxygen Species (ROS) in tobacco cells treated with BABA 24 hours prior to the addition of each elicitor. Further, tyrosine phosphorylation of SIPK kinase residues was determined by Western Blot together with the expression of selected genes involved in defense response. Finally, plasma membrane isolation was done to measure proteome changes by LC-MS/MS analysis. The results showed a significant increase in ROS production after BABA treatment in all studied elicitors. However, this increase seems to be not related to pre-activation of signaling pathways or increased expression of defense genes induced by BABA treatment. The main factor responsible for this increase could be higher concentration of proteins responsible for the perception of elicitors on the plasma membrane. However, due to time reasons, complex proteomic analysis of isolated tobacco plasma membranes has not been finished.
