Abstract:

Použití cytostatik na bázi komplexů těžkých kovů je jednou z možností léčby nádorových onemocnění v současné lékařské praxi. Přes všechny klady této léčby je jejich použití limitováno některými faktory, mezi které patří například nežádoucí výskyt rezistence buněk nádorové tkáně vůči používanému cytostatiku či nadměrná zátěž pacienta vedlejšími účinky cytostatika. Hlavní mechanizmy ovlivnění biologických struktur neionizujícím ultrazvukovým polem jsou spojeny zejména s jevy způsobenými prostupujícím mechanickým vlněním. Mezi hlavní faktory, ovlivňující živé soustavy v souvislosti s ultrazvukovým ozvučením, je tepelný ohřev, kavitace a mechanické síly. Předložená disertační práce shrnuje výsledky studia kombinovaného působení ultrazvukového pole o nízkých intenzitách a cytostatika cisplatiny na buněčné linie in vitro. Cílem disertační práce bylo vyhodnotit vliv těchto fyzikálně-chemických faktorů v závislosti na jejich experimentálním uspořádání na viabilitu buněk linií lidského ovariálního karcinomu A2780 a A2780cis. Výchozím předpokladem pro vznik této experimentální práce byl známý fakt účinku ultrazvukového pole na buněčné struktury, zejména možnost ovlivnění struktur buněčných membrán. Tento účinek ultrazvukového pole se jeví jako možný způsob, kterým lze docílit zvýšeného příjmu cytostaticky aktivních látek do nitra buňky. Z provedených experimentů a naměřených dat je zřejmé, že viabilitu daných buněčných linií A2780 a A2780cis ovlivňuje nejen samostatně podané cytostatikum cisplatina, ale zejména byla ovlivněna viabilita experimentálních skupin buněk vystavených kombinovanému působení ultrazvukového pole s cisplatinou. Výsledky ukazují, že přítomnost cytostatika v živném mediu v průběhu ozvučení způsobuje větší pokles viability, oproti experimentu, kdy bylo cytostatikum přidáno až po provedeném ozvučení. Na základě provedených experimentů je možné také usoudit, že k ovlivnění buněčných povrchů dochází již během průběhu procesu ozvučení a jejich určitá reparace nastává bezprostředně po ukončení ozvučení. Získané výsledky naznačují možnou cestu ke zvýšení účinnosti chemoterapie nádorových onemocnění s využitím ultrazvukového pole o nízkých intenzitách.

Abstract:

Using of antitumor complexes of heavy metals is one of the treatment options for cancer in current medicine. Despite all the positive aspects of this therapy, some less favorable factors still need to be considered, for example an occurrence of side resistance of tumor cells to chemotherapeutic agents or side effects of drugs causing neuropathy, nephropathy, nausea etc. The main mechanisms of non-ionizing ultrasound field effect on biological structures are especially associated with the phenomena caused by mechanical waves. The main factors affecting the biological system in the context of ultrasonic sound system is thermal heating, cavitations and mechanical stress. This dissertation summarizes the results of studies of combined action of ultrasonic fields of low intensities and cytostatics cisplatin on cell lines in vitro. The aim of this thesis was to evaluate the influence of physicochemical factors of the experimental arrangement of the cell viability of human ovarian cancer lines A2780 and A2780cis. The underlying assumption for the formation of this experimental work was a known fact of the effect of ultrasonic field on the cell structure, particularly the possibility of affecting the structures of cell membranes. The effect of ultrasonic field appears to be a conceivable method, which can be achieved by increasing intake of cytostatically active agents to the cells core. It was shown, that viability of the cell lines A2780 and A2780cis affects not only the separate administration of cytostatically drug cisplatin, but that the viability of the cell lines was particularly impacted by the experimental groups of cells which have been exposed to the combined action of ultrasound field with cisplatin. The results show that the presence of cytostatics in the cell culture medium during the sonication has resulted in a greater decrease of cells viability, compared to the experiment when the cytostatics was added after sonication. Based on the experiments carried out, it can also be concluded that the effect of altered cell surface occurs during the process of ultrasound applications; however, some self-recovery occurs immediately after the ultrasound exposure. The gathered data suggest another potential method, presenting a feasible improvement of increasing the effectiveness of cancer chemotherapy by using ultrasound fields of low intensities.
