Study of host-pathogen interactions on the chicken-Salmonella model

Abstract:

Salmonella enterica subsp. enterica je jedním z nejčastějších původců gastroenteritid u lidí, přičemž nejběžnějším zdrojem tohoto patogenu jsou vejce a drůbeží maso. Vakcinace drůbeže, která by vedla ke snížení kontaminace chovů salmonelami, je proto považována za účinné opatření ke snížení incidence salmonelóz v lidské populaci. Většina dostupných vakcín byla vytvořena chemickou nebo UV mutagenezí a jejich molekulární podstata tudíž není dobře charakterizována. V našich studiích jsme proto testovali vakcinační potenciál dvou kmenů se sníženou virulencí, delečních mutantů S. enterica sv. Enteritidis postrádajících ostrov patogenity SPI1 nebo SPI2. Vakcinace mutantem SPI1 chránila kuřata před infekcí divokým kmenem lépe, vakcinace SPI2 mutantem oproti tomu vedla k vyšším hladinám protilátek. Vakcinace mutantem Salmonella Enteritidis ΔSPI1 navíc chránila kuřata také před infekcí serovarem Typhimurium. Z praktických důvodů byl připraven vakcinační kmen postrádající flagelin Salmonella Enteritidis ΔSPI1 Δlon ΔfliC s mukoidním vzhledem. Chovy drůbeže vakcinované touto vakcínou budou snadno odlišitelné od chovů infikovaných terénními kmeny díky tomu, že vakcinační kmen u kuřat nevyvolává tvorbu protilátek proti flagelinu. Druhá část práce je věnována obecné odpovědi kuřat na infekci Salmonella Enteritidis ve střevě a slezině, tedy oblasti, která je zatím až na cytokinovou a protilátkovou odpověď nepříliš známa. Čerstvě vylíhlá kuřata reagovala na infekci salmonelou signalizací pomocí cytokinů IL-8 a IL-17, produkcí antibakteriální proteinu lysozymu g2 a interferonem-indukovaného proteinu IFIT5. Matrix metalloproteináza 7, enzym štěpící složky mezibuněčného prostoru, rozvolnila tkáň a umožnila tak prostup heterofilů a makrofágů k lumen střeva. IFN-gamma spolu s dalšími cytokiny aktivoval transkripční regulátory STAT1 a STAT3, což vedlo k diferenciaci Th1 a Th17 lymfocytů. Šestitýdenní nevakcinovaná kuřata odpovídala na infekci S. Enteritidis silnou prozánětlivou reakcí charakterizovanou především Th17 lymfocyty produkujícími IL-17, zatímco vakcinovaná kuřata reagovala produkcí IFN-gamma zprostředkovanou Th1 buňkami. Produkce avidinu, sérového amyloidu A, trappinu 6, chemokinu AH221, lipocalinu ExFABP a proteinu IRG1 zřejmě hraje v odpovědi kuřat na salmonelu významnou roli. Exprese těchto genů byla ve střevě naivních kuřat po infekci mnohonásobně zvýšena, zatímco u vakcinovaných kuřat se jejich exprese po infekci nezměnila. Tyto geny tedy mohou sloužit jako markery odpovědi kuřat na infekci salmonelou.

Abstract:

Salmonella enterica subsp. enterica is one of the most abundant causative agents of gastroenteritis in human, with eggs and poultry meat being the main sources of this pathogen. Vaccination of poultry, which would decrease Salmonella contamination of flocks, is therefore considered to be an effective measure to reduce salmonellosis cases in human. Most of the current vaccines against salmonellosis were prepared by chemical or UV mutagenesis and their molecular bases are not characterized well. In our studies we therefore tested a vaccine potential of two strains attenuated in their virulence, Salmonella pathogenicity island (SPI) 1 and SPI2 deletion mutants of S. enterica serovar Enteritidis. Vaccination with SPI1 deletion mutant was shown to be more protective, while vaccination with SPI2 mutant led to higher antibody levels in chickens. Vaccination with SPI1 mutant of Salmonella Enteritidis protected chickens against infection with Salmonella Typhimurium. For practical purposes, a triple mutant vaccine Salmonella Enteritidis ΔSPI1 Δlon ΔfliC, with mucoid phenotype was constructed. Poultry flocks vaccinated with this vaccine will be easily distinguishable from infected flocks, as this vaccination strain fails to induce production of anti-flagellin antibodies. As Jim Kaufman once said, “Chickens are not mice with feathers.” In our next studies we therefore focused on the general response of chickens to Salmonella Enteritidis in the intestine, the field which is not explored in detail except for cytokine and antibody response. Newly hatched chickens responded to S. Enteritidis infection with IL-8 and IL-17 signaling, antibacterial protein lysozyme g2 and interferon inducible protein IFIT5. Matrix metalloproteinase 7, enzyme degrading components of extracellular matrix, allowed tissue relaxation with subsequent heterophil and macrophage translocation to the intestinal lumen. Later on, IFN-gamma and other cytokines activated transcriptional regulators STAT1 and STAT3 mediating differentiation of Th1 and Th17. Six week old non-vaccinated chickens responded to S. Enteritidis infection with a strong proinflammatory response characterized by Th17 cells producing IL-17, while vaccinated chickens responded to the infection with IFN-gamma produced by Th1 cells. Apart from cytokines, avidin, serum amyloid A, trappin-6, chemokine AH221, lipocalin ExFABP and immune responsive gene 1 were shown to be central to the control of chicken response to S. Enteritidis infection. As the transcription of these genes increased in the cecum of naïve chickens after the infection, but essentially did not change in vaccinated chickens, these acute response genes may be used as new markers for chicken response to Salmonella infection.
