Study of Antimicrobial Activity of Biopolymer Mixtures

Abstract:

Diplomová práce se zaměřuje na zkoumání antimikrobiální aktivity polymerních směsí. V teoretické části jsou blíže specifikovány základní nosné systémy zahrnující emulze, emulgely, lipozomy, micely, či polymerní částice. V této práci byly připraveny biopolymerní směsi zein/chitosan s přídavkem antimikrobiálních látek (thymol, esenciální oleje - tymián, skořice a oregano), které byly charakterizovány (zeta potenciál, pH, průměrná velikost částic), vizuálně zkoumány, byly sledovány jejich antimikrobiální účinky a také dynamika uvolňování aktivní látky. Roztoky ve vyšších koncentracích u thymolu, tymiánu a oregana působili na gramnegativní bakterie. Naopak roztoky s vyšším obsahem skořice vytvářeli větší inhibiční zóny u grampozitivní bakterie a působili na rozdíl od předchozích i na plíseň. Všechny vzorky obsahující 2% thymol vykazovaly inhibiční účinky. V další části práce byly vytvořeny filmy, jako nejpovedenější se jevily s minimálním obsahem zeinu 2 %. Z výsledků bylo patrné, že filmy měly mnohem větší antimikrobiální aktivitu a to především u plísně Aspergillus niger, na kterou roztoky vůbec neúčinkovali. Nakonec byla provedena studie zaměřená na uvolňování aktivních látek v čase. Většina vzorků po 24 hodinách již nevykazovala žádné inhibiční účinky.

Abstract:

The diploma thesis focuses on the investigation of the antimicrobial activity of polymer mixtures. The theoretical part concerns about the basic carrier systems including emulsions, liposomes, micelles, and polymer particles. In this work, biopolymer mixtures of zein/chitosan were prepared with addition of antimicrobial compound (thymol, essential oils - thyme, cinnamon, or oregano). These solutions were characterized (zeta potential, pH, average particle size), and visually observed. What more, antimicrobial activity and dynamics of active substance release, were determinated. Solutions in higher concentrations of thymol, thyme and oregano acted on gram-negative bacteria. The advantages of solutions with higher shell values are larger inhibition zones for gram-positive bacteria and an effect on small and medium money. All types of 2% thymol equipment showed inhibitory effects. In other parts of the work, films were evaluated, as the most successful appeared with a minimum zein rating of 2%. The results showed that the films have greater greater antimicrobial potential and cause the fungus Aspergillus niger, they did not work at all to solve the problems. Finally, a study focused on the release of active substances over time was performed. The other samples no longer showed any inhibitory effects after 24 hours.
