Study of drug release from matrix tablets based on polyvinyl alcohol

Abstract:

Tato práce je zaměřena na studium vlivu obsahu retardující komponenty na uvolňování účinné látky z matricových tablet. Jako modelová účinná látka byla použita kyselina salicylová. Vliv retardující složky byl studován v hydrofilních systémech na bázi polyvinylakoholu (Parteck? SRP 80) a lipofilních systémech na bázi glyceryl dibehenátu (Compritol? 888 ATO). Tablety byly připraveny metodou přímého lisování. Zjištěné závěry byly získány na základě disolučních testů a nelineární regresní analýzy. Bylo zjištěno, že v systémech na bázi polyvinylakoholu pojivo neovlivňuje ani mechanismus, ani rychlost uvolňování kyseliny salicylové. Také bylo zjištěno, že rychlost uvolňování klesá s rostoucím obsahem retardantu. U systémů na bázi glyceryl dibehenátu také klesala rychlost uvolňování kyseliny salicylové s rostoucím obsahem retardační složky. Formulace s obsahem 5 % glyceryl dibehenátu vykazovala odchylky od předpokládaného chování. Mechanismus uvolňování léčiva z hydrofilních a lipofilních tablet je rozdílný.

Abstract:

The aim of this thesis is to study the effect of retarding component content on the release of active pharamaceutical substance (API) from matrix systems. Salicylic acid was used as model API. The effect of the retarding component was studied in polyvinyl alcohol hydrophilic systems (Parteck? SRP 80) and lipophilic systems based on glyceryl dibehenate (Compritol? 888 ATO). The tablets were prepared by the compression method. Results were obtained based on dissolution tests and nonlinear regression analysis. The binder does not have effect to release and release mechanism of the API. It has also been found that the release rate decreases with increasing retardant content. For glyceryl dibehenate systems, the release rate of salicylic acid also decreased with the increasing content of the retardant component. Formulations containing 5% glyceryl dibehenate showed deviations from predicted behavior. The mechanism of release from hydrophilic and lipophilic tablets is different.
