Effect of water pH on the quality of lavender oil obtained by hydrodistillation

Abstract:

Tato diplomová práce se zabývá levandulovým esenciálním olejem. První část teoretické části je věnována samotné rostlině, je zde popsán především její výskyt, vzhled a vlastnosti. Další část je zaměřena obecně na esenciální oleje. Jsou zde uvedeny vlastnosti olejů a také metody, kterými se esenciální oleje nejčastěji získávají. Dále se tato část zabývá chemickým složením esenciálních olejů, kde je popsána především hlavní skupina látek, ze kterých se esenciální oleje skládají. Touto hlavní skupinou jsou terpeny. Obvykle bývají hlavní dvě nebo tři složky zastoupeny v poměrně vysokých koncentracích (20?70 %) ve srovnání s ostatními složkami, které jsou přítomny pouze ve stopových množstvích. Poslední kapitola teoretické části je věnována analýze esenciálních olejů. Identifikace jednotlivých složek esenciálních olejů a stanovení jejich relativního zastoupení se obvykle provádí pomocí plynové chromatografie. Nejčastěji ve spojení s hmotnostním spektrometrem (GC-MS) a pomocí plynového chromatografu vybaveného plamenovým ionizačním detektorem (GC-FID). Dále jsou v této kapitole uvedeny metody pro stanovení biologických vlastností esenciálních olejů, konkrétně se jedná o stanovení antimikrobiální a antioxidační aktivity. V experimentální části práce je zkoumán vliv pH vody na kvalitu esenciálních olejů získaných hydrodestilací. Tato část je také zaměřena na stanovení antimikrobiální aktivity vydestilovaných esenciálních olejů.

Abstract:

This diploma thesis deals with lavender essential oil. The first part of the theoretical part is focused on the plant itself. It describes mainly its occurrence, appearance and properties. The next part is focused on essential oils in general. The properties of the oils are given here, as well as the methods by which the essential oils are gradually obtained. Furthermore, this part deals with the chemical composition of essential oils. The main group of substances of which essential oils are composed is described here. Terpenes are the main group. Usually, the main two or three components are present in different high concentrations (20?70%) compared to other components, which are present only in trace amounts. The last chapter of the theoretical part is devoted to the analysis of essential oils. The identification of the individual components of the essential oils and the determination of their relative proportions are usually carried out by gas chromatography. Most often in conjunction with a mass spectrometer (GC-MS) and a gas chromatograph equipped with a flame ionization detector (GC-FID). Furthermore, this chapter presents methods for determining the biological properties of essential oils, specifically the determination of antimicrobial and antioxidant activity. The experimental part of the work investigates the effect of water pH on the quality of essential oils obtained by hydrodistillation. This part is also focused on determining the antimicrobial activity of distilled essential oils.
