Using the transgenic DR5::GFP line to study apical dominance in Arabidopsis thaliana (L.)

Abstract:

Tato bakalářská práce zkoumala protokol a praktickou implementaci techniky projasňování vzorků rostlinných pletiv, které exprimují gen pro zelený fluorescenční protein (GFP) jako reportérový systém pod auxinem indukovatelným promotorem DR5. V teoretické části práce byly rozebrány hlavní aspekty problematiky využiti fluorescenčních proteinů (převážně GFP). Zde jsou popsány také apikální dominance (AD) a její význam v zemědělskou praxi, role fytohormonů,zejména auxinu, v regulace AD. Popsán je také promotor DR5, jeho použití v současných studiích a shrnutí problematiky projasňování vzorků pletiv a tkání pro mikroskopii. V rámci praktické části této práce pak byly testovány různé protokoly projasňování vzorků za účelem pozorování fluorescence GFP. Výsledky experimentů byly diskutovány se závěry jiných prací, zaměřených na tuto problematiku.

Abstract:

The protocol and practical implementation of tissue clearing techniques in plant tissues expressing the gene for green fluorescent protein (GFP) as a reporter system under the auxin-inducible promoter DR5 were investigated in this bachelor thesis. The main aspects of utilizing fluorescent proteins (mainly GFP) for studying gene expression were examined in theoretical part. Apical dominance (AD) and its potential applications in agricultural practices, the role of phytohormones in AD regulation, particularly auxin have also been studied within the scope of the research. Furthermore, the paper has addressed the DR5 promoter, its recent applications in studies, and summarized tissue clearing methods for microscopic observation. In the practical part of this work, different protocols of fixation and clearing of the samples were tested. The results of the experiments were compared with other works on this subject.
