Characterization of molecular markers responsible for non-specific resistnace in plants

Abstract:

V přírodě rostliny neustále musí čelit patogenním mikroorganismům, mezi ně například patří houby, oomycety, viry, bakterie nebo nematody. Aby byly schopny přežít ve svém prostředí, vyvinuly si vlastní indukovatelné obranné mechanismy, které jsou dosud předmětem intenzivního výzkumu. K výzkumu mechanismů nespecifické rezistence rostlin, byl v této práci použit modelový systém sestávající z hostitelské rostliny Nicotiana tabacum a elicitinu kryptogeinu vylučovaného patogenní oomycetou Phytophtora cryptogea. V první části je na základě získaných výsledků nastíněna možná role specifických lysinových zbytků v biologické aktivitě elicitinů. Výsledky dále naznačují možnou interakci elicitinů s endogenním partnerem v rostlině, nejpravděpodobnějším kandidátem jsou proteiny přenášející lipidy. Druhá část práce je věnována proteomické a transkriptomické analýze proteinů asociovaných s plazmatickou membránou, které se zapojují do obranné reakce elicitované kryptogeinem. Získané znalosti by mohly být dále využity při studiu markerů jiných, ekonomicky významných rostlin.

Abstract:

In the nature, plants are constantly under attack of pathogenic microorganisms such as fungi, oomycetes, viruses, bacteria or nematodes. To be able to survive in their environment they have therefore evolved their own inducible defence mechanisms which are still intensively researched. In order to get an insight into non-specific resistance in plants, a model system consisting of a host plant Nicotiana tabacum and elicitin cryptogein secreted by pathogenic oomycetes Phytophtora cryptogea was used in this work. In the first part, based on obtained results, the role of specific lysine residues in the elicitins’ biological activity is suggested. Moreover, a possible interaction of elicitins with another endogenous partner in plant, the most likely candidate being the lipid transfer proteins, is indicated. The second part is dedicated to proteomic and transcriptomic analysis of tobacco plasma membrane-associated proteins involved in defense responses triggered by cryptogein. The acquired knowledge might be further used in the study of markers of other economically important plants.
