Determination of molar mass and molar mass distribution of polysaccharides using size exclusion chromatography

Abstract:

Bakalářská práce se zabývá stanovením molární hmotnosti a distribuce molárních hmotností polysacharidů pomocí gelové permeační chromatografie. Úvodní část práce pojednává o významu polysacharidů, věnuje se jejich struktuře, vlastnostem a rozdělení. Další část práce je zaměřena na vybraného zástupce polysacharidů {--} škrob, který je fyziologicky a technologicky nejvýznamnějším polysacharidem. Druhá část práce je věnována gelové permeační chromatografii jako metodě, pomocí níž lze stanovit molární hmotnost a distribuci molárních hmotností polysacharidů. Tato kapitola shrnuje poznatky o kolonách, detektorech a kalibraci používaných v gelové permeační chromatografii. Poslední kapitola se věnuje konkrétním aplikacím gelové permeační chromatografie při stanovení molárních hmotností a jejich distribuce u polysacharidů.

Abstract:

This bachelor thesis deals with determination of molar mass and molar mass distribution of polysaccharides using size exclusion chromatography. In the introduction, importance of polysaccharides simultaneously with focus on their structure, properties and classification is mentioned. The next part of the study is directed to one of the polysaccharide representatives {--} starch, which is physiologically and technologically the most important polysaccharide. In the second part of the theses, size exclusion chromatography, as a method for determination of molar mass and molar mass distribution of polysaccharides is described. This chapter summarizes information about columns, detectors and calibration used in size exclusion chromatography. In the last part of this work, applications of size exclusion chromatography for polysaccharides analyses are resumed.
