Abstract:

Práce se zabývá testováním vsádkového typu čipového mikrokalorimetru nazývaného také IC-kalorimetr. Přístroj byl vyvinut na Technické Univerzitě ve Freibergu Dr. Lerchnerem. Jedná se o typ kalorimetru, který není komerčně dostupný, a proto bylo nutné provést řadu experimentů, které přispějí k detailnějšímu popisu a k dokonalejší znalosti zařízení, ke spolehlivému měření a vyhodnocování výsledků a nahradí tak absenci dostupné literatury. Hlavní měřící jednotkou kalorimetru je komerčně dostupný čip NCM-9924 dodávaný firmou Xensor Integration. Charakteristickou veličinou popisující každý čip je citlivost, kterou lze považovat za kalibrační konstantu. Studovanými parametry ovlivňujícími citlivost čipu, přesnost a správnost měření byly teplota, objem reakční směsi, intenzita vlhčení kroužku, individuálnost čipu, temperační doba a tloušťka membrány. Bylo zjištěno, že na stabilitu výstupního signálu mají největší vliv charakter látky na čipu a teplota. Získané poznatky byly využity ke studiu tří enzymatických reakcí: ureázy s močovinou, invertázy se sacharózou a trypsinu s a N?-benzoyl-L-arginin-p-nitroanilid hydrochloridem. U těchto reakcí byly stanoveny reakční entalpie za různých podmínek a studovány vlivy koncentrací enzymu a substrátu, pH, složení pufrů a teploty na rychlost reakcí. Všechny uvedené faktory významně ovlivňují rychlost enzymatických reakcí.

Abstract:

The work deals with testing of the batch type chip microcalorimeter otherwise known as IC-calorimeter. Calorimeter was developed at the Technical University of Freiberg by Dr. Lerchner. This type of calorimeter is not commercially available and it was necessary to make a lot of experiments that would contribute to more detailed description and complete knowledge of the equipment, to reliable measurement and evaluation of results and replace the lack of available literature. The main measuring segment of the calorimeter is a commercially available chip NCM-9924 supplied by Xensor Integration. The characteristic quantity describing of each chip is the sensitivity, which can be regarded as a calibration constant. Temperature, reaction mixture volume, intensity of wetting of the ring, the individuality of the chip, tempering time and thickness of the membrane were the studied parameters affecting sensitivity of the chip and accuracy of measurements. It was found that the character of the substance and the temperature have the greatest influence on the output signal stability.The gained findings were utilized to studying of three enzymatic reactions: the reaction of urease with urea, sucrose with invertase and trypsin with the N?-benzoyl-L-arginine-p-nitroanilide hydrochloride. Enthalpy of these reactions under different conditions was determined and effects of enzyme and substrate concentrations, pH, buffer composition and temperature on reaction rate were studied. All these factors significantly influence the rate of enzymatic reactions.
