Genetic engineering in bioleaching microorganisms

Abstract:

Bioloužení jako způsob získávání nerostných materiálů je rozvíjející se odvětví biotechnologií představující alternativní způsob těžby surovin po celém světě. V zásadě se jedná o využití metabolismu mikroorganismů k uvolnění a následné extrakci chtěné látky ze sloučeniny či směsi. Toto většinou probíhá za silně kyselých podmínek, což je pro většinu organismů neslučitelné se životem. Protože genetické inženýrství bylo ohlášeno jen pro málo zástupců acidofilních mikroorganismů, shrnuje tato práce dosavadní pokrok v genetickém inženýrství dvou bioloužících mikrobiálních rodů, ve kterých bylo dosaženo na tomto poli největších výsledků. První je bakteriální rod Acidithiobacillus, který patří k nejpoužívanějším a nejlépe prozkoumaným organismům tohoto odvětví. Zejména díky využití plasmidu s širokým rozsahem hostitelů, pJRD215, bylo na úrovni kyvadlových vektorů pro Acidithiobacillus dosaženo mnohých úspěchů. Druhým rodem je hypertermofilní acidofilní archea Sulfolobus, u kterého dosavadní výzkum naznačuje vysoký bioloužící potenciál v budoucích průmyslových aplikacích.

Abstract:

Bioleaching as a means of obtaining mineral materials is an evolving field of biotechnology, representing an alternative way of mining used worldwide. It involves exploiting of microorganism metabolism to release and extract the desirable substance from a chemical compound or mixture. This usually takes place in extremely acidic conditions that are not compatible with life for most of the organisms. Because genetic engineering has been reported only for a few of the acidophilic microorganisms, this thesis summarizes only the progress in genetic engineering of two microbial bioleaching genera in which the best results have been achieved. The first one is a bacterial genus Acidithiobacullus, which has been one of the most used and best-investigated organisms in this field. Especially thanks to the use of a plasmid with a broad range of hosts, pJRD215, many achievements in the field of shuttle vectors for Acidithiobacillus have been made. The second one is a hyperthermophilic acidophilic archaeon Sulfolobus. The research shows very promising results that indicate the potential for industrial use of this genus.
