Detekce plísně bramboru a rajčete pomocí zobrazovací fluorescence chlorofylu, infračerveného termálního zobrazování a hyperspektrálního zobrazování

Detection of late blight of potato and tomato by chlorophyll fluorescence imaging and infrared thermal imaging and hyperspectral imaging

Abstract:

Hlavním cílem této práce bylo vypracování literární rešerše zabývající se detekcí houbových chorob lilku bramboru a plísně rajčete s využitím zobrazovacích metod, provedení analýzy jejich výhod a nedostatků a na vzorcích listů s variabilní intenzitou napadení experimentem vyhodnotit metody detekce hyperspektrálního zobrazování, zobrazování UV stínění fluorescence, metody zobrazovací fluorescence chlorofylu a infračerveného termálního zobrazování. V rámci experimentu byla provedena detekce plísně bramboru a následně ke vzorkům listu bramboru přidán rostlinný materiál lilku rajčete, který díky větší škále napadení poskytoval vhodné doplnění analýzy využitelnosti zobrazovacích metod pro detekci napadení chorobami. Z výsledků vyplývá skutečnost, že všechny zkoumané metody vykazují možnosti využití v rámci detekce napadení patogenem P. infestans. Průkazná korelace pro závislost k napadení byla zjištěna pro maximální kvantový výtěžek fotosystému II (FV/FM). Infračervené termální zobrazování se i přes svou účinnost neosvědčilo jako spolehlivá metoda pro predikci napadení z důvodu neprůkazné závislosti. Metoda UV stínění fluorescence, která byla použita vykazovala vysokou průkaznost pro plíseň bramboru. Díky této metodě lze konstatovat závislost mezi mírou napadení plísně bramboru a akumulací flavonoidů v epidermálních buňkách listu. Stejné metody byly v experimentu použity také na rostlinný materiál rajčete. Byla objevena podobnost charakteru interakcí patogenu a hostitelské rostliny s obdobnou fyziologickou odezvou mezi oběma druhy rostlin. Infračervené termální zobrazování vykazovalo u rajčete s významnou průkaznou závislost k napadení, ovšem menší ve srovnání s ostatními metodami. Metoda hyperspektrálního zobrazování u obou druhů vykazovala významné hodnoty spolehlivosti predikce napadení. Je zřejmý potenciál využití zkoumaných metod detekce napadení houbových chorob ať už pro monitoring, fenotypizaci, prognózu apod. Rozhodujícím faktorem budou požadavky jednotlivých metod, vzhledem k množství zkoumaného rostlinného materiálu, času potřebného pro detekci, potřebné citlivosti nebo vzdálenost senzorů od listů.

Abstract:

The main goal of this thesis was to write a literature review of the detection of potato fungal diseases and late blight of tomato using imaging methods, analysis of their advantages and disadvantages and on samples of leaves with variable phase of attack experimentally evaluate methods of hyperspectral imaging, UV fluorescence imaging and chlorophyll fluorescence imaging. In the experiment, the detection of potato blight was found and subsequently the plant material of tomato was added to the potato leaf samples, assuming the occurrence of a common pathogen. The results show that all investigated methods show potential uses in the detection of P. infestans. The UV fluorescence shielding method used on the potato leaf showed high evidence. Thanks to this method, it is possible to state the dependence between the degree of infestation of potato blight and the accumulation of flavonoids in leaf epidermal cells. The quantum yield of photosystem II of the chlorophyll fluorescence method also achieved conclusive results, when a close dependence of the pathogen infestation level on the measured parameters was found. Infrared thermal imaging, despite its effectiveness, has not proven to be a reliable method for predicting infestations due to inconclusive dependence. The same methods were also used in the experiment on tomato plant material. A similarity in the nature of the pathogen-host plant interactions with a similar physiological response was found between the two plant species. Infrared thermal imaging was significantly demonstrable in tomato, but smaller compared to other methods. The method of hyperspectral imaging in both species showed significant values of reliability of prediction of infestation. There is an obvious potential for the use of researched methods of fungal disease detection for monitoring, prognosis, etc. The decisive factor will be the requirements of individual methods, given the amount of plant material, time required for detection, required sensitivity or distance of sensors from leaves.
