Pharmacological modulation of the nitric oxide production in the process of inflammation

Abstract:

Oxid dusnatý (NO) je molekula s celou řadou důležitých fyziologických funkcí, mezi něž patří relaxace cévní hladké svaloviny, přenos nervového vzruchu, dále hraje úlohu při agregaci krevních destiček a v imunitním systému. Právě NO produkovaný makrofágy hraje klíčovou roli v nespecifické imunitě jako významný mikrobicidní faktor proti intracelulárním bakteriím nebo vlastním poškozeným buňkám. Při jeho nadprodukci je však schopen toxicky působit i vůči vlastním tkáním. Zvýšená produkce NO je rovněž typická pro některá chronická onemocnění jako astma nebo revmatoidní artritidu. Některé studie naznačily, že NO hraje významnou úlohu i v alergickém zánětu. NO je schopen inhibibovat aktivaci žírných buněk, ale zároveň oplývá i řadou prozánětlivých účinků. Práce byla zaměřena především na vliv vybraných H1-antihistaminik (10 léků 1.generace a 4 léky 2. generace) a dále carvedilolu, léku ze skupiny blokátorů β-adrenergních receptorů, na produkci NO makrofágy in vitro, zejména vliv těchto léků na expresi inducibilní syntázy NO (iNOS) a produkci a akumulaci nitritů, konečného produktu metabolismu NO. Jako vhodný buněčný model pro experimenty byla použita linie myších makrofágů RAW 264.7. Přímé vychytávací schopnosti H1-antihistaminik a carvedilolu vůči NO byly studovány v nebuněčném systému pomocí amperometrické analýzy. Pro určení lipofility zkoumaných H1-antihistaminik bylo využito chromatografické metody. Po 24 hodinách inkubace s buňkami, H1-antihistaminika v netoxických koncentracích inhibovala expresi proteinu iNOS a akumulaci nitritů, což ovšem nebylo způsobeno přímými vychytávacími schopnostmi H1-antihistaminik. Úroveň inhibice akumulace nitritů pozitivně korelovala s lipofilitou testovaných látek. Z výsledků vyplývá, že modulace produkce NO by mohla být způsobena snížením exprese iNOS proteinu a/nebo aktivity tohoto enzymu. Carvedilol v testovaných koncentracích inhiboval expresi iNOS proteinu a snižoval koncentraci nitritů, ovšem jeho přímé vychytávací schopnosti vůči NO rovněž nebyly prokázány.

Abstract:

NO is an agent with a plenty of important physiological functions, such as smooth muscle relaxation, neurotransmission, platelet aggregation or immune mechanisms. NO produced by macrophages plays a crucial role in the innate immunity as an important microbicidal factor in the fight against intracellular bacterias or tumor cells. However, the overproduction of NO causes the toxicity also against tissues of host organism and is also obvious for chronic inflammatory diseases such as asthma or rheumatoid arthritis. Nevertheless recent studies observed, that NO also plays an important role in allergic reactions. NO is able to inhibit mast cells activation, but also has proinflammatory effects. The thesis is mainly focused on the effects of selected H1-antihistamines (10 drugs of the 1st generation and 4 drugs of the 2nd generation) and carvedilol, drug from the group of β-adrenergic receptors, on the NO production by macrophages in vitro, especially the effect of these drugs on the expression of inducible nitric oxide synthase (iNOS) responsible for the NO generation and then the effect of these drugs on the accumulation of nitrites, the end product of NO metabolism. As the convenient model for the experiments were used murine macrophage cell line RAW 264.7. The direct scavenging properties of H1-antihistamines and carvedilol against NO were studied amperometrically in the cell-free system. For the lipophilicity parameter of tested H1-antihistamines determination was used chromatographical assay. After a 24 hour incubation with cells, H1-antihistamines in pharmacological concentrations inhibited the iNOS protein expression and nitrite accumulation. This decrease in nitrite accumulation was not caused by the scavenging ability of drugs against NO. The degree of inhibition of the nitrite accumulation positively correlated with the degree of tested H1-antihistamines lipophilicity. In conclusion, the modulation of nitric oxide production could be caused by the downregulation of iNOS protein expression and/or the iNOS protein activity. Carvedilol inhibited iNOS protein expression and decreased the nitrite concentration in cell supernatants, however its direct scavenging properties aganist NO were not proved.
