Cytostatic activity of organometallic complexes containing V, Nb and Mo

Abstract:

V diplomové práci bylo provedeno studium inhibičního účinku 66 organokovových komplexů s centrálním atomem V, Nb a Mo. Testování bylo provedeno na nádorových liniích buněk MOLT-4 a HL-60. Jako vysoce cytotoxické pro obě linie buněk byly shledány komplexy vanadu a molybdenu s N,N´- chelátově vázaným fenantrolinem a jeho deriváty. Na vysoce účiném komplexu vanadu (V16) a molybdenu (Mo13) byl studován mechanizmus cytotoxického účinku. Pomocí průtokové cytometrie, spektrálních metod a Western blottingu v kombinaci SDS-PAGE s imunochemickou detekcí bylo prokázáno, že oba dva testované komplexy indukují apoptózu prostřednictvím aktivace vnitřní apoptické dráhy u linie buněk MOLT-4 a Jurkat. U buněčné linie MOLT-4 bylo prokázáno zvýšení exprese tumor supresorového genu, proteinu p53 a jeho fosforylované formy na serinu 15 po působení V16 i Mo13.

Abstract:

In the diploma thesis, a study of the inhibition effect of 66 organometalic complexes with V, Nb and Mo as a central atom was made. The tests were done on the cancer cell lines MOLT-4 and HL-60. The complexes of V and Mo with the chelate-bounded phenantroline and its derivates were determined as highly cytotoxic for both cell lines. The mecanism of the cytotoxic effect was studied on the highly efficient vanadium (V16) and molybdenum (Mo13). The induction of the apoptosis via an internal apoptic way activation at the MOLT-4 and Jurkat cell lines was established for both complexes with the flow cytometry, spectral methods and the Western blotting in a combination with the SDS-PAGE and an imunochemical detection. At the MOLT-4 cell line, an elevation of the expression of the tumor supressor gene, protein p53 and its fosforylated form on serin 15 was prooved for both V16 and Mo13.
