The role of hypoxia in myeloid differentiation

Abstract:

Ačkoliv je úloha hypoxie u solidních nádorů široce studovaným tématem, studií možných účinků hypoxie na leukemické buňky existuje zatím jen velmi omezené množství. Na základě současných poznatků o možném spojení mezi procesem diferenciace leukemických buněk a hypoxií nebo mimetik hypoxie, bylo stanoveno za hlavní cíl této práce analyzovat a srovnat účinek hypoxie, modelových mimetik hypoxie a standardně používaných diferenciačních činidel na diferenciaci leukemických linií HL60, NB4 a U937. K ověření diferenciačního statusu buněk byla v této práci použita například analýza expresního profilu specifických transkripčních faktorů pomocí metody qRT-PCR nebo fenotypizace buněk detekcí specifických povrchových antigenů pomocí průtokové cytometrie. Z výsledků práce vyplývá, že buněčná odpověď na působení použitých činidel a reálné hypoxie je značně heterogenní v závislosti na stádiu diferenciace, ve kterém se jednotlivé linie nacházejí. Zároveň se však potvrdilo, že hypoxie i její mimetika regulují diferenciaci pozitivním směrem.

Abstract:

Although the role of hypoxia in solid tumors is a widely studied topic, there is still very limited number of studies of possible effects of hypoxia on leukemia cells. Based on current knowledge suggesting a possible link between leukemic cell differentiation and hypoxia or hypoxia mimetics, the main aim of this thesis was to analyze and compare the effect of hypoxia, model hypoxia mimetics and standard differentiation agents on differentiation of leukemic cell lines HL60, NB4 and U937. In order to verify the differentiation status of the cells, for example the analysis of the specific transcription factor profiles using the qRT-PCR method or the detection of specific surface markers by flow cytometry phenotyping were used in this thesis. The results of this thesis show that the cellular response to the effects of the real hypoxia and hypoxia mimetics is considerably heterogeneous, depending on the differentiation stage of the individual cell lines. At the same time, however, it has been confirmed that hypoxia and its mimetics regulate differentiation in the positive direction.
