The study on AGR2 protein role in pancreatic cell lines

Abstract:

Protein AGR2 náleží do tzv. rodiny anterior gradient proteinů. Bylo prokázáno, že nadměrná tvorba nebo naopak potlačení exprese tohoto proteinu může ovlivňovat buněčnou proliferaci, invazivitu, citlivost k léčbě, metastázování a růst nádorů in vitro i in vivo. V nedávné době se také ukázalo, že zvýšená exprese AGR2 může u karcinomu pankreatu ovlivňovat citlivost na gemcitabin, což je hlavní lék, který se podává při léčbě tohoto onemocnění. Karcinom slinivky patří mezi smrtelné choroby s celosvětově stoupající incidencí a velice špatnou prognózou, u kterého není v současné době používaná terapie ani zdaleka tak účinná jako u jiných typů nádorů. Tato práce se zabývala studiem úlohy drah, které regulují AGR2 a skrze které by mohla být snížena hladina tohoto proteinu, což by mohlo vést ke zlepšení citlivosti při léčbě gemcitabinem. Zjistili jsme, že pro snížení hladiny AGR2 je nejefektivnější inhibice dráhy EGFR, MAPK a mTOR. Při současné inhibici těchto drah byl pozorován synergistický účinek, který vedl k výraznému potlačení hladiny tohoto proteinu. Funkční analýza s využitím MTT testu pak potvrdila, že kombinovaná aplikace gemcitabinu společně s inhibicí signálních drah regulujících hladinu AGR2 významně umocňuje cytotoxický účinek. Výsledky této práce naznačují možnost budoucího využití tohoto proteinu jako prediktivního faktoru pro zefektivnění léčby karcinomu pankreatu.

Abstract:

AGR2 protein belongs to the anterior gradient proteins family. It was proved that overexpression or conversely suppression of this protein may have an impact on cellular proliferation, invasiveness, sensitivity to the treatment, metastasizing and tumor growth both in vitro and in vivo. It was recently shown that an increased expression of AGR2 may, in case of a pancreatic carcinoma, influence sensitivity to gemcitabine which represents the treatment of choice for this disease. Pancreatic cancer is a common, highly lethal disease that is rising in incidence with poor outcome. Current therapy is not so efficient in comparison with other types of tumors. This thesis was focused on the role of pathways that are involved in regulation of AGR2. We speculate that inhibition of these pathways may improve sensitivity to gemcitabine treatment. We found that the inhibition of the EGFR, MAPK and mTOR pathways is the most effective for decreasing of AGR2 level. A synergistic effect which led to a substantial suppression of AGR2 level was observed during simultaneous inhibition of these pathways. A functional analysis using the MTT test then confirmed that combined application of gemcitabine with the inhibition of signal pathways that regulate the level of AGR2 significantly enhances the cytotoxic effect. Results of this work suggest a possible future utilization of this protein as a predictive factor for streamlining the treatment of pancreatic carcinoma.
