Analýza exprese kandidátních biomarkerů spojených s rezistencí k retinoidům u buněk neuroblastomu

Expression analysis of candidate biomarkers of resistance to retinoids in neuroblastoma cells

Abstract:

Neuroblastom je maligní nádor dětského věku. Ve své nejtěžší formě vyžaduje agresivní léčbu, při které nalezly významné uplatnění retinoidy. Jedním z problémů při použití retinoidů je ovšem vznik rezistence. Možným řešením tohoto problému je zlepšení predikce odpovědi pacienta na léčbu a to s pomocí vhodného prediktivního biomarkeru. Cílem této diplomové práce byla analýza exprese genů vybraných jako vhodné kandidátní markery u buněčných linií derivovaných z neuroblastomu. Hlavní část práce je zaměřena na změny u sbírkových linií SK-N-BE(2) a SH-SY5Y derivovaných z neuroblastomu po ovlivnění syntetickými i přírodními retinoidy. Sledovány byly změny v expresi vybraných biomarkerů, jejich lokalizace v buňce a také růstové parametry. Dílčí část práce se věnuje analýze exprese vybraných biomarkerů na úrovni mRNA u unikátního souboru 21 pacientských buněčných linií. Práce prokázala senzitivitu obou sbírkových linií k použitým retinoidům. U obou linií způsobovaly retinoidy snížení proliferace, na druhou stranu měly jen nepatrný vliv na buněčný cyklus. V rámci sledování buněčného cyklu byl sledován i podíl apoptózy a zatímco u linie SK-N-BE(2) byl patrný nárůst podílu apoptotických buněk u linie SH-SY5Y byl pozorován spíše pokles. Při srovnání různých typů retinoidů se projevily jen minimální rozdíly v jejich působení, zejména v ovlivnění růstových parametrů, některé výraznější rozdíly byly zaznamenány při analýze změn exprese konkrétních genů. Ze sledovaných potenciálních markerů se jako nejzajímavější jeví geny HOXC9 a PBX1, u kterých byla exprese po ovlivnění retinoidy výrazně vyšší oproti neovlivněné kontrole. Další výzkum těchto genů může být přínosem při studiu mechanismu rezistence. U genů DDX39A, HMGA1 a NF1 nebyla zjištěna přímá souvislost s působením retinoidů ani s rezistencí k retinoidům. Zjištěno naopak bylo, že nedetekovatelná exprese HOXC9 a naopak prokazatelná exprese HMGA2 může souviset s rezistencí k retinoidům.

Abstract:

Neuroblastoma is a children’s malignant tumor. In its the most difficult form, it requires aggressive treatment that includes use of retinoids. One problem with retinoids is resistance. A possible solution to this problem is to improve the prediction of the patient's response to the treatment, using a suitable predictive biomarker. The aim of this diploma thesis was to analyze expression of genes selected as suitable candidate markers in neuroblastoma derived cell lines. The main part of the work is focused on changes in the collection lines SK-N-BE (2) and SH-SY5Y derived from neuroblastoma after the influencing with synthetic and natural retinoids. Changes in the expression of selected biomarkers, their localization in the cell, and growth parameters were also observed. The partial part deals with the analysis of the expression of selected biomarkers at mRNA level in a unique set of 21 patient cell lines. We demonstrated the sensitivity of both collection lines to the retinoids. On both lines, retinoids caused decreasion of proliferation, on the other hand, they had only a slight effect on the cell cycle. The proportion of apoptosis was also monitored as part of the cell cycle observation, and whereas the SK-N-BE (2) line showed an increase in the proportion of apoptotic cells at the SH-SY5Y line it was decrease. Comparison of the different types of retinoids did not reveal differences in their effects, especially in influencing growth parameters, some more significant differences were noted in the analysis of changes in expression of particular genes. The most interesting potential markers are HOXC9 and PBX1 genes, where the expression after the retinoid effect was significantly higher than in the uninfluenced control. Further research on these genes may be helpful in studying the resistance mechanism. In the DDX39A, HMGA1 and NF1 genes, no direct relationship to effect of retinoids or retinoid resistance was found. It was found that the undetectable expression of HOXC9 and detectable expression of HMGA2 may be related with resistance to retinoids.
