Prevalence of Apicomplexa and Spirochaetes representatives in vectors and hosts

Abstract:

Zoonotické patogeny představují závažné zdravotní riziko. Tato práce se zaměřuje na čtyři patogenní mikroorganismy, jejichž vliv na zdravotní stav jak člověka, tak zvířat může být vážný – Babesia sp., Toxoplasma gondii, Leptospira sp. a Borreliella burgdorferi sensu lato. V teoretické části bude čtenář obeznámen se všemi výše zmíněnými mikroorganismy, stejně jako s taxonem Ixodiae. Zástupci tohoto taxonu jsou vektory některých uvedených patogenů, u ostatních uvedených patogenů se takový přenos uvažuje. Experimentální část této diplomové práce shrnuje detaily provedeného výzkumu. Byl proveden sběr klíšťat a jiných savčích hematofágních roztočů, metodou PCR byl zjištěn výskyt DNA uvedených patogenů ve volné přírodě uvnitř jejich prokázaných i potenciálních vektorů. Dále byli odchyceni divoce žijící malí savci, metodou ELISA a MAT jim byly vyšetřeny séra a výplachy srdcí na protilátky proti třem z těchto patogenů. Metodou PCR byl potvrzen výskyt DNA všech uvedených patogenů v tělech odchycených klíšťat, nejčastější byla Borreliella burgdorferi s. l. Promoření rezervoárních populací savců na všech lokalitách patogeny Toxoplasma gondii, Leptospira sp. a Borreliella burgdorferi sensu lato bylo potvrzeno.

Abstract:

Zoonotic patogens pose a serious health risk. This thesis focuses on four pathogenic microorganisms, which may greatly affect health conditions of people as well as animals – Babesia sp., Toxoplasma gondii, Leptospira sp. and Borreliella burgdorferi sensu lato. In the theoretical part, the reader will be apprised with all of the previously mentioned microorganisms as well as with the Ixodidae taxon. Members of this taxon are vectors of some of the mentioned pathogens , transmission of the other mentioned pathogens is considered. The experimental part of this thesis summarises the details of the conducted research. Ticks and other haematophagous mites were collected, occurence of DNA of the aforementioned microorganisms in their proven or potential vectors was detected by the PCR method. Furthermore, small wild mammals were caught, their sera and heart rinses were examined for antibodies against three of the four pathogens by ELISA and MAT methods. Occurence of DNA of all of the previously mentioned pathogens in bodies of collected ticks was proven, most abundant pathogen was Borreliella burgdorferi s. l. Immunity rate of the reservoar populations of mammals on all sites by Toxoplasma gondii, Leptospira sp. and Borreliella burgdorferi sensu lato was proven.
