Detection of viruses in grapevines infected by of GTD pathogens

Abstract:

GTD (Choroby kmínku révy vinné z angl. „Grapevine trunk diseases“) patogeny představují závažnou hrozbu pro révu vinnou (Vitis vinifera L.), avšak v certifikačním schématu není jejich výskyt sledován. V certifikačním schématu je obsaženo pouze 6 povinně testovaných virů. Vzhledem k těmto faktům byla vytvořena tato studie pro zmapování koinfekcí mezi těmito patogeny. Konkrétně bylo metodou RT-PCR detekováno 10 virů (včetně 6 virů spadajících do certifikačního schématu) a 2 viroidy. Byly testovány 4 vinohrady z oblasti Pálava, ze kterých bylo odebráno 40 vzorků rostlin, ve kterých byly v rámci projektu TAČR Zéta 2 č. TJ02000096 detekovány rody GTD patogenů. K řešené problematice doposud nebyla publikována žádná studie. Výsledky dokumentují frekvenci výskytu virů a viroidů v oblasti Pálava v rostlinách napadených GTD patogeny a dále možné ovlivnění mezi zkoumanými rostlinnými patogeny. Práce by měla být rozšířena za účelem hlubšího porozumění tématu koinfekce GTD patogenů a virů.

Abstract:

GTD (Grapevine trunk diseases) pathogens cause serious damage of grapevines (Vitis vinifera L.) nevertheless their incidence is not controlled by the certification scheme. Only a six obligatorily tested viruses are included in the certification scheme. Due to these factors, this study focuses on mapping of coinfection between GTD and viruses and viroids. There were detected 10 viruses (including 6 viruses pertain to the certification scheme) and two viroids by RT-PCR. Four vineyards from Palava region were surveyed, 40 samples of grapevines were sampled based on GTD symptoms occurence, fungal detection was done within a project TAČR Zéta 2 No. TJ02000096. No study has been published regarding this experimental topic before. The results show the frequency of viruses and viroids occurrence in Palava region in plants infected by GTD pathogens and the possible influencing between those various groups of pathogens. The experimental topic has to be extended to better understand to the coinfections because the subject of the experiment is huge.
