Biological active metabolites of plants in the family Amaryllidaceae

Abstract:

V současné době se inhibitory cholinesteráz řadí mezi nejúčinnější léčiva, která zmírňují projevy Alzheimerovy choroby a dalších neurodegenerativních onemocnění s cholinergním deficitem. Proto hledání nových efektivnějších inhibitorů cholinesteráz patří mezi hlavní cíle řady studií. Tato diplomová práce se zabývá sekundárními metabolity rostlin čeledi Amaryllidaceae, konkrétně druhu \kur{Nerine Bowdenii} W. Watson, a jejich izolací, identifikací a biologickou aktivitou. Vlastní izolace alkaloidů z rostlinného extraktu byla provedena zejména pomocí tenkovrstvé chromatografie a následnou GC/MS a NMR analýzou byla zjištěna struktura těchto látek. Izolované alkaloidy byly podrobeny testování jejich inhibiční aktivity vůči acetylcholinesteráze a butyrylcholinesteráze pomocí Ellmanovy metody. Účinnost inhibitorů je uváděna prostřednictvím hodnoty IC50. Dále byl změřen rozdělovací koeficient izolovaných alkaloidů v soustavě 1-oktanol:voda pro zjištění prostupnosti studovaných látek přes hematoencefalickou bariéru in vitro.

Abstract:

Inhibitors of cholinesterases are currently one of the most effective drugs to alleviate symptoms of Alzheimer's disease and other neurodegenerative disorders with a cholinergic deficit. Therefore the search for new and more effective inhibitors of cholinesterases is a major goal for many studies. This thesis deals with secondary metabolites of plants in the family Amaryllidaceae, in particular Nerine Bowdenii W. Watson species, and their isolation, identification and biological activity. The actual isolation of alkaloids from a plant extract was performed mainly by thin layer chromatography, followed by GC/MS and NMR analysis, which indicated the structure of these substances. Isolated alkaloids were subjected to inhibitory activity testing against acetylcholinesterase and butyrylcholinesterase using Ellman's method. The efficacy of inhibitors is presented in IC50 value. Further, the partition coefficient of isolated alkaloids in a system 1-octanol:water was measured to determine the permeability of the studied substances through the blood-brain barrier \kur{in vitro}.
