Influence of different methods on seed germation Sarracenia

Abstract:

Teoretická část se zabývá charakteristikou rodu Sarracenia, uvedením do problematiky klíčení semen, dormance a vlivu abiotických faktorů. Praktická část se zabývá (1) kvalitativním stanovením podílu semen Sarracenia (plných, prázdných, napadených hmyzem a semen mechanicky poškozených) v rozborovém vzorku pomocí RTG zkoušky a také (2) kvantitativním vyhodnocením pozorování klíčení semen Sarracenia u různých metod předosevní přípravy (stratifikace, stimulace fytohormonem GA3, skarifikace pomocí H2SO4 a skarifikace pomocí Sava). Práce obsahuje pracovní postupy předosevní přípravy semen, nasbíraná data z jednotlivých pozorování klíčení semen, analýzu dat, jejich vyhodnocení a následný popis. Experimenty probíhaly v období 2012/2013. Cílem práce bylo zjistit vliv metod předosevní přípravy na klíčivost semen u vybraných druhů Sarracenia. Testované vzorky mezi sebou porovnat a získat metodu, která vykazuje nejvyšší klíčivost semen Sarracenia.

Abstract:

The theoretical part deals with the characteristics of the genus Sarracenia, introduction to the issue of seed germination, dormancy and the influence of abiotic factors. The practical part deals with (1) the qualitative determination of the proportions of seeds Sarracenia (full, empty, affected by insects and seeds mechanically damaged) in the sample analyzed by the RTG (radiographic) examination, and (2) the quantitative evaluation of the Sarracenia seed germination observation using different pre-sowing preparation methods (stratification, stimulating fytohormone GA3, scarification with H2SO4 and scarification using Savo). The work includes the techniques of pre-sowing seeds preparation, collected data from individual observations of seed germination, data analysis, evaluation and subsequent description. The experiment was conducted in the period 2012/2013. The aim of this work was to determine the effect of pre-sowing preparation methods on seed germination of selected species of Sarracenia. Furthermore, to compare the test samples with each other and to get a method which shows the highest seed germination of Sarracenia.
