Effect of oxidative stress on proteome of bioleaching bacteria

Abstract:

This thesis deals with qualitative changes in proteome of bioleaching bacterium Acidithiobacillus ferrooxidans under the oxidative stress caused by hydrogen peroxide, which occurs naturally in the biomining environment and biogeochemical processes in sulphide deposits. An introduction of the thesis is an overview of knowledge about bioleaching bacteria, specifically about a model microorganism At. ferrooxidans and application possibilities. The thesis was focused on metabolism of At. ferrooxidans and its role in oxidation. Second part of the introduction deals with oxidative stress and its effects on the cells and their defense and relation between Fenton reaction and oxidative stress, which affects bacteria living in the metal rich environment. Earlier literature proteomic results related to stressed bacteria are summarized at the end. A kinetic inhibitory parameter after the induction of the oxidative stress in At. ferrooxidans using hydrogen peroxide was specified. A label-free qualitative MS-based proteomics was used to study an oxidative stress in At. ferrooxidans CCM 4253. Proteomic response to a hydrogen peroxide-induced oxidative stress revealed a wide spectrum of proteins involved in various metabolic pathways.

Abstract:

Tato práce se zabývá kvalitativními změnami v proteomu bioloužící bakterie Acidithiobacillus ferrooxidans při oxidativním stresu způsobeným peroxidem vodíku. Ten se může přirozeně vyskytovat v prostředí, kde dochází k biotěžbě kovů nebo spontánním biogeochemickým procesům v prostředí sulfidových minerálů. Teoretická část poskytuje souhrn poznatků o bakterii At. ferrooxidans a její vztah k oxidaci pyritu. Dále se úvod do problematiky zabývá oxidativním stresem a jeho vlivem na buňky a jejich následnou obranu. Je diskutován vztah mezi Fentonovou reakcí a oxidativním stresem, který působí zejména na organismy žijící v prostředí bohatém na kovy. Závěrem jsou shrnuty dosavadní literární poznatky o vlivu stresu na proteom studovaných bakterií. V praktické části byl stanoven kinetický parametr inhibice po působení oxidativního stresu na bakterii At. ferrooxidans, který byl vyvolán peroxidem vodíku. Na studium oxidativního stresu v At. ferrooxidans CCM 4253 byla využita nezávislá kvalitativní MS analýza proteomu. Proteomická odpověď na stres indukovaný peroxidem vodíku odhalila široké spektrum proteinů zahrnutých v různých metabolických dráhách.
