Mechanisms of endocrine resistance in breast cancer

Abstract:

This bachelor thesis deals with a study of biomarkers of resistance to endocrine therapy and its mechanisms. The theoretical part includes the literature review focused on the classification of breast cancer and on types of endocrine therapy. It further describes the mechanisms of resistance to endocrine therapy of breast cancer including the role of previously identified biomarkers. The experimental part of this thesis is dedicated to the interpretation of results of the proteomic analysis of endometrial carcinomas. Associations between levels of proteins and previous tamoxifen therapy, tumor grade and invasiveness were identified in SWATH proteomics analysis. The results are discussed.

Abstract:

Předložená bakalářská práce se věnuje studiu biomarkerů rezistence vůči endokrinní terapii a jejími mechanismy. Teoretická část práce zahrnuje literární rešerši zaměřující se na klasifikaci nádorů prsu a typy endokrinní terapie. Dále popisuje mechanismy rezistence vůči endokrinní terapii u nádorů prsu včetně úlohy dosud identifikovaných biomakerů. V experimentální části se práce zabývá interpretací výsledků proteomické analýzy nádorů endometria. Analýzou proteomických SWATH dat byly zjišteny vztahy mezi hladinami proteinů a předchozí terapií tamoxifenem, nádorovým grade a invazivitou. Výsledky analýzy jsou diskutovány na základě rozsáhlé literární rešerše.
