Isolation and detection methods of foodborne viruses from food and environmental matrices

Abstract:

Tato práce je věnovaná problematice izolačních a detekčních metod virů, které zapříčiňují alimentární infekce. I když k šíření těchto virů dochází nejčastěji prostřednictvím mezilidského kontaktu, má svůj výrazný podíl na jejich šíření i přenos prostřednictvím kontaminovaných potravin. Správný odhad rizika, které tyto kontaminace představují, ale znesnadňuje fakt, že testování potravin na přítomnost virových kontaminací není běžným postupem při jejich výrobě, zpracování a distribuci. Celkové riziko těchto kontaminací a následně vzniklých infekcí tak může být výrazně podhodnoceno. Svoji roli zde hraje i nedostatek standardizovaných postupů pro izolaci a posléze detekci virů alimentárních infekcí z daných potravin. Různé přístupy jsou navíc často úzce aplikovatelné a proto je zapotřebí optimalizovat metody pro danou matrici a konkrétní virové agens. Tato práce je proto zaměřena na přehled nejpoužívanějších metod pro vhodnou izolaci a detekci virových částic z různých potravinových matric a z prostředí, ve kterém se mohou tyto potraviny vyskytnout.

Abstract:

This thesis is focused on the isolation and the detection methods of the viruses that cause food-borne infections. Although the spread of these viruses occurs most often through interpersonal contact, transmission via contaminated food has a significant role on transmission. But the correct estimation of the risk is difficult due to the fact that testing of foodstuff for the presence of viral contamination during its growing, processing and distribution is not a common practice. Therefore the overall risk of the contaminations and subsequently the outbreaks can be significantly underestimated. A lack of standardized methods for isolation and detection of foodborne viruses plays its role too. Various approaches are often narrowly applicable, and thus it is necessary to optimise the method for the specific matrix and viral agens. Therefore, the aim of this thesis is summarisation of the most commonly used methods for appropriate isolation and detection of viral particles from different food matrices and the environment in which this foodstuff may appear.
