Grapevine defence gene expression in the field

Abstract:

Vinná réva ve stresových situacích aktivuje komplex vlastních obranných mechanismů. Během obranné reakce dochází ke zvýšení exprese obranných genů. Tyto obranné geny mají svůj podíl na aktivní obraně rostliny vůči patogenům. Pomocí kvantitativní PCR byly sledovány rozdíly v expresi genů v listech vinné révy, a to u genů Glub, Chib, Chic a VVTL, které kódují proteiny pro basickou glukanasu, basickou isoformu chitinasy, kyselou isoformu chitinasy a vitis vinifera thaumatin-like protein. Rozdíly v expresi byly sledovány v období od konce června do poloviny srpna, kdy rostliny byly postupně vystaveny infekčnímu tlaku plísně révové, padlí révového a plísně šedé. Během infekčního tlaku se výrazně měnila exprese těchto obranných genů v sledovaných rostlinách. K největšímu nárůstu došlo v poslední fázi odběru od 13.7 do 12.8, kdy exprese genů vzrostla až deset tisíc krát. Nejmenší rozdíly byly naměřeny v expresi genu Glub a Chib u rostliny 2. Byly pozorovány rozdíly v expresi jednotlivých genů při odběru z jedné rostliny a vzájemné rozdíly mezi třemi rostlinami. Největší variabilita v expresi byla prokázána u genu Glub. Exprese tohoto genu se ve sledovaných rostlinách vyvíjela velmi rozdílně. Pro rostlinu č. 2 byla exprese téměř konstantní, u rostlin č. 1 a 3 došlo nejdříve v polovině července k poklesu exprese, po kterém, v období v polovině srpna, následoval nárust exprese, jenž byl u rostliny č. 3 velmi výrazný. Naopak gen Chic se projevil jako gen s nejméně variabilní expresí. Průběh jeho exprese se ve studovaných rostlinách během jednoho a půl měsíce příliš neměnil. Jeho exprese byla téměř homogenní. U genů Chic a VVTL se exprese u jednotlivých rostlin také příliš nelišila, ale naopak se výrazně měnil její průběh s rostoucím působením infekčního tlaku houbových patogenů během studovaného období. Dále byla sledována celková variabilita obranných genů vrámci jedné rostliny a variabilita mezi více rostlinami. Bylo prokázáno, že variabilita vrámci jedné rostliny není kupodivu menší než variabilita mezi rostlinami.

Abstract:

Grapevine is activating various complex of defense mechanisms in stress times. There is increase of and expression of defensive genes during the defensive reaction. These genes have taken deal during the active defense against the pathogens attack. We were observing differences in genes expression in the grapevine leaves using quantitative PCR . Genes of Glub, Chib, Chic and VVTL, which codes proteins for alkaline glucanase, alkaline isoform of chitinase, acidic isoform of chitinase and vitis vinifera thaumatin-like protein, were observed. Differences in expression were monitored from the end of June till the middle of August. During this period the plants were gradually exposed to infection pressure of Plasmopara viticola, Uncinula necator and Botrytis cinerea. During the infection attack there were significant change of expression of these defensive genes in studied plants. The largest increasment was at the last phases of the monitoring between 13.7. and 12.8., when the expression raised almost ten-thousand times more. The smallest differences were measured in genes Glub and Chib expression on the plant No.2. We saw differences in the expression of individual genes in samples from one plant and mutual differences between three tested plants. Greatest variability in the expression was proven in Glub gene. The expression of this gene in tested plants showed various results for each plant during monitoring period. The expression was almost constant on plant No.2, plants No.1 and No.3 showed decreasing of the expression till the half of July then before the middle of August increasement was seen, on plant No.3 this increasement was very significant. Gene Chic on the other hand demonstrate itself with lowest expression variability. The course of his expression in tested plants was almost stable with only small changes during test period of one and half Month. His expression was almost homogenous. There was no so much difference in the expression of Chic and VVTL genes on the individual plants, but there were significant changes of expression progression caused by raised influence of fungal pathogens during tested period. We were observing whole variability of defensive genes in one plant and variability of genes between plants. It was proven, that variability of one individual plant is not smaller than variability of couple of plants.
