Roles of cytochrome C in the cell

Abstract:

Bakalářská práce se zabývá cytochromem c a jeho funkcí v eukaryotické buňce. V úvodu práce je popsána mitochondrie, její struktura a funkce. Další kapitoly jsou zaměřeny na elektron transportní řetězec a jeho jednotlivé komplexy jako jsou cytochrom c reduktáza/oxidáza a samotný cytochrom c. U cytochromu c je popsána jeho struktura, role v oxidační fosforylaci, stejně tak jako u cytochrom c reduktázy a oxidázy. Dále je popsána jeho konformační dynamika, antioxidační účinky, účast během apoptózy, a nakonec jeho interakce s toxiny. Vysvětleny jsou základní principy přenosu elektronů cytochromem c v dýchacím řetězci a vznik kyslíkových radikálů v mitochondrii. Velká pozornost je věnována účasti cytochromu c v apoptóze buňky. Podrobně je popsán mechanismus aktivace apoptózy, dále uvolnění cytochromu c z mitochondrie a tvorba apoptozomu. V závěru práce je objasněn princip ovlivnění cytochromu c a celého dýchacího řetězce toxiny.

Abstract:

The bachelor thesis deals with cytochrome c and its function in eukaryotic cells. The introduction describes mitochondria, its structure and function. Following chapters are focused on electron transport chain and respiratory complexes like cytochrome c reductase/oxidase and finally cytochrome c. The structure of cytochrome c as well as of cytochrome c reductase and oxidase is described including roles in oxidative phosphorylation. In addition, conformation dynamics of cytochrome c, antioxidant effects, participation in apoptosis and modulation of its activity by toxins are described. In the bachelor thesis, basic principles of electron transfer by cytochrome c in the respiratory chain are explained including reduction of oxygen radicals in mitochondria. Great attention is paid for participation of cytochrome c in apoptosis. The activation mechanism of apoptosis is described in detail, i.e. release cytochrome c from mitochondria and formation of apoptosome.
