Detection of Interspecific Gene Flow in the Genus Cirsium

Abstract:

Tato bakalářská práce obsahuje literární rešerši k detekci genového toku pomocí různých molekulárních markerů (haplotypů chloroplastové DNA, mikrosatelitů a polymorfizmu délek amplifikovaných fragmentů) a jejich dosavadního využití v rodu Cirsium (pcháč). Na tuto teoretickou část pak navazuje část praktická, která je zaměřena na studium principu genového toku v rámci středoevropských pcháčů a testování jeho detekce pomocí různých druhů molekulárních markerů. K tomuto účelu byly ve třech různých oblastech (Brno a okolí, Bílé Karpaty a Žďárské vrchy) vzorkovány tři skupiny druhů lišící se mírou hybridizace: často hybridizující (C. oleraceum, C. palustre, C. rivulare), hybridizující omezeně (C. acaule, C. canum, C. heterophyllum), nehybridizující nebo hybridizující jen velmi vzácně (C. arvense, C. eriophorum, C. vulgare). Práce je zaměřena převážně na výběr druhů a oblastí sběru pro experiment. Dále pak dokumentuje výsledky testování všech tří metod, na základě kterých byla jako vhodný marker zvolena metoda AFLP, pomocí které bude genový tok dále analyzován v navazující magisterské práci.

Abstract:

This Bachelor´s thesis contains a literature review on detection of gene flow using various molecular markers (chloroplast DNA haplotypes, microsatellites and amplified fragment lenght polymorphism) and their current use in the genus Cirsium (thistle). The theoretical part is followed by a practical part, which is focused on a study of the gene flow pattern in Central European thistles and the testing of its detection with the aid of different sorts of molecular markers. For this purpose a sampling in three different regions (Brno and its surroundings, White Carpathians and Žďárské vrchy hills) of three groups of species: often hybridizing (C. oleraceum, C. palustre, C. rivulare), restrictidly hybridizing: (C. acaule, C. canum, C. heterophyllum) and non-hybridizing or very rarely hybridizing: (C. arvense, C. eriophorum, C. vulgare). This thesis is focused mainly on the choice of species and localities used for this experiment. Further, it documents provisional analyses, on the basis of which the suitable marker (AFLP) was chosen. With the use of AFLP maker the data will be further analysed in the subsequent Master´s thesis.
