Glycoluril derivatives and its dimerization

Abstract:

Glykoluril a zejména jeho deriváty se pro zakřivení své struktury a možnost tvorby vodíkových vazeb ukazují jako vhodné stavební bloky větších supramolekulárních objektů. Z těch se pro výjimečné hostitelské vlastnosti asi největší pozornosti dostává především jeho makrocyklickým oligomerům, tzv. cucurbiturilům. Ve snaze porozumět procesu, kterým se tyto strukturně složité molekuly vytváří, a tak potenciálně vyřešit problém jejich nízké rozpustnosti nalezením obecně úspěšné metody syntézy jejich derivátů, byly jako modelový systém první fáze jejich tvorby zkoumány dimerizační reakce chráněných glykolurilů a následná izomerizace vznikajících diastereomerů. Tato diplomová práce se konkrétně zaměřuje na studium izomerizace S-dimeru 2,8-dimethylglykolurilu v prostředí kyseliny HCl, která byla na rozdíl od dosud studovaných reakcí zvolena jako systém respektující reálné podmínky syntézy cucurbiturilových struktur. S využitím metod NMR byla tato reakce charakterizována jak z kinetického, tak termodynamického hlediska a taktéž byl proveden experiment poukazující na intermolekulární průběh této přeměny. Dále byly vedle této studie též syntetizovány nové deriváty glykolurilu, a to 1-methyl-4,6-o-xylylenglykoluril a jeho cyklický ether, které byly následně využity jako prekurzory pro syntézu dimerních a trimerních struktur. Taktéž je popsána syntéza 2,8-dimethylglykolurilu a jeho S-dimeru, u nějž pak byla právě sledována jeho izomerizace na termodynamicky stabilnější C-dimer.

Abstract:

Glycoluril and especially its derivatives are with the curvature of their structures and ability to form hydrogen bonds ones of the suitable building blocks for larger supramolecular objects. Of those the most attention is mainly given to its macrocyclic oligomers, host molecules which are called cucurbiturils. In an effort to understand the process via these structural difficult molecules are formed and thus potentially solve the problem of their poor solubility by invention of generally working method of the synthesis of their derivatives were investigated as the model system of the first stage of their formation the dimerization reactions of the protected glycolurils and consecutive isomerizations of the generated diastereomers. This diploma thesis is namely focused on the isomerization of the S-dimer of 2,8-dimethylglycoluril in the presence of hydrochloric acid which was contrary to formerly studied reactions chosen with respect to the real conditions of the cucurbituril synthesis. This reaction was characterized by NMR methods and was determined both its kinetic and thermodynamic parameters. Furthermore the cross-over experiment revealed that this conversion is intermolecular process. In addition to this study was also synthesized new derivatives of glycoluril, 1-methyl-4,6-o-xylylenglykoluril and its cyclic ether, which were subsequently used as the precursors for the synthesis of the dimer and trimer glycoluril structures. There is also described the synthesis of 2,8-dimethylglycoluril and its S-dimer whose isomerization to the more thermodynamic stable C-dimer was just studied.
