Importance of myeloperoxidase in regulation of cell death of neutrophil granulocytes

Abstract:

Zánětlivý proces, spojený s aktivací neutrofilních granulocytů, umožňuje organismu bojovat s invazí patogenů. Avšak jeho přetrvávání vede k neustálé infiltraci neutrofilních granulocytů, které mohou trvale poškodit tkáň, a proto musí být z místa zánětu včas odstraněny nastoupením programované buněčné smrti. Po aktivaci oxidativního vzplanutí dochází u neutrofilních granulocytů k expresi fosfatidylserinu (z anglického phosphatidylserine, PS) na povrchu buněk, což je typický znak pro apoptózu. Dřívější studie ukazují, že exprese PS je snížená u neutrofilních granulocytů, které jsou deficientní na myeloperoxidázu (MPO). MPO je tedy pravděpodobně významně zapojena v regulaci programované buněčné smrti. Tato diplomová práce studuje vliv MPO na regulaci a mechanismus buněčné smrti u neutrofilních granulocytů po stimulaci aktivátory oxidativního vzplanutí: phorbol 12-myristát 13-acetátem (PMA), opsonizovanou bakterií Streptococcus mutans (OST) a N-formylmethionin-leucyl-fenylalaninem (fMLP). Byl prokázán účinek aktivátorů na produkci reaktivních metabolitů kyslíku pomocí luminolem amplifikované chemiluminiscence. Dle očekávání byl zjištěn velmi vysoký rozdíl v chemiluminiscenci u kontrolních a MPO deficientních neutrofilních granulocytů. Inhibitor MPO, 4-ABAH, vykazoval pouze limitovaný efekt na chemiluminiscenci neutrofilních granulocytů aktivovaných PMA a OST. Detekcí exprese PS byla prokázána nižší frekvence buněčné smrti aktivovaných neutrofilních granulocytů deficientních na MPO. Naopak přidáním externí MPO k deficientním buňkám došlo k poklesu životnosti na úroveň kontrolních neutrofilních granulocytů. Navzdory expresi PS nebyla pozorována aktivace dalších apoptotických ukazatelů (proteiny BID a BAX, aktivace kaspázy 3 a 8). Naopak byla pozorována aktivace autofagických proteinů (p62 a LC3) a také nekroptotických ukazatelů (RIP1 a MLKL). Byla rovněž zaznamenána citrulinace histonu H3, která úzce souvisí s NETózou, a také citrulinace histonu H3 v kombinaci s expresí PS, avšak bez významného rozdílu mezi kontrolními a MPO deficientními neutrofilními granulocyty. Celkově získaná data podporují hypotézu významného vlivu MPO na průběh buněčné smrti u neutrofilních granulocytů, ale bude potřeba bližší charakterizace daného tématu.

Abstract:

Acute inflammation connected with migration of neutrophil granulocytes into the site of inflammation allows organism to deal with a disruption of host integrity. However, prolonged infiltration of neutrophil granulocytes into the inflamed tissue can cause irreversible damage. Therefore, their removal from inflammatory site by programmed cell death is necessary. Earlier studies showed lower externalization of phosphatidylserine (PS) on the surface of myeloperoxidase (MPO)-deficient neutrophil granulocytes. Therefore, MPO seems to be significantly involved in a regulation of this process. This master thesis is focused on the role of MPO in a regulation of programmed cell death of neutrophil granulocytes stimulated by activators of oxidative burst: phorbol 12-myristate 13-acetate (PMA), opsonized Streptococcus mutans (OST) and N-formylmethionine-leucyl-phenylalanine (fMLP). Ability of selected activators to induce reactive oxygen species production was proved by using luminol-amplified chemiluminescence. As expected, significantly lower signal was observed in neutrophil granulocytes isolated from MPO-deficient mice. Inhibitor of MPO, 4-ABAH, showed only limited impact on chemiluminescence of neutrophil granulocytes activated by PMA and OST. Detection of PS confirmed delayed cell death of activated MPO-deficient neutrophil granulocytes. On the other hand, an external addition of MPO to MPO-deficient granulocytes mediated a decrease in viability of MPO-deficient cells to the levels of cells isolated from control mice. However, despite the increased PS expression of activated neutrophil granulocytes, other markers of apoptosis were not found to be activated (proteins BID and BAX, caspase 3 and 8). Changes in expression of autophagic proteins (p62 and LC3) and necroptotic markers (RIP1 and MLKL) were observed. Also, histone H3 citrulination, which is closely related to NETosis, as well as histone H3 citrulination in combination with PS expression were observed. But, there was not a significant difference between control and MPO deficient neutrophil granulocytes. Overall, the obtained data support the hypothesis of a significant effect of MPO on the course of cell death in neutrophil granulocytes. However, future studies are needed for more detailed characterization of this phenomenon.
