Determination of reference values of matrix metalloproteinases 2, 3, 9 and asymmetric dimethylarginine

Abstract:

Práce je věnována problematice matrixových metaloproteináz se zaměřením na matrixové metaloproteinázy 2, 3, 9 a asymetrického dimethylargininu, metodám stanovení těchto markerů a stanovení jejich referenčních intervalů. Matrixové metaloproteinázy jsou proteolytické enzymy podílející se na štěpení bílkovin extracelulární matrix. Jejich význam je možné spatřovat např. při regeneraci tkání, hojení, zánětu. Asymetrický dimethylarginin vzniká při metabolizaci bílkovin z argininu působením enzymů methyltransferáz. Přirozeně se vyskytuje v lidském organismu a významně brání tvorbě oxidu dusnatého. Volba zaměření konkrétně na tyto markery vychází z jejich provázanosti s kardiovaskulárními onemocněními, stenózami a in stent restenózami.

Abstract:

The work is dedicated to the problems of the matrix metalloproteinases focusing on matrix metalloproteinases 2, 3, 9 and asymmetric dimethylarginine, the methods of these markers determination and the determination of their reference intervals. The matrix metalloproteinases are the proteolytic enzymes participating on degrading of the extracellular matrix proteins. Their significance can be seen e.g. in tissue repair, healing, inflammation. Asymetric dimethylarginine arises at the process of the proteins metabolism from arginine due to infuence of methyltransferase enzymes. He naturally occurs in the human body and impedes to nitrogen oxide production. Selecting focus specifically on these markers is based on their connection with the cardiovascular diseases, stenosis and restenosis in stent.
