Cannabis sativa - clarifying the relationship between the putative subspecies

Abstract:

Tato diplomová práce prezentuje problematiku klasifikace a definice dvou domnělých taxonů konopí, sativa a indica a uvádí výsledky komplexní studie rozmanitosti kultivarů konopí získaných jako reprezentanti typů sativa a indica. Hypotéza o existenci chemických a genetických parametrů pro rozlišení dvou domnělých poddruhů sativa a indica je testována metodami pro zjišťování chemického a genetického profilu. Konkrétně se jedná o plynovou chromatografii a analýzu SNP metodou KASP firmy LGC. V rámci chemické analýzy bylo metodou GC-MS u 191 analyzovaných vzorků Cannabis identifikováno celkem 21 monoterpenů, 19 seskviterpenů a 8 kanabinoidů. Osm terpenů bylo u konopí popsáno nově. Multivariantní model analýzy hlavních komponent umoţnil odhalit význam terpenů pro určení typů sativa a indica od dodavatele, a byly tak popsány potenciální biochemické markery pro jejich rozlišení. Izolací DNA metodou CTAB byla získána vysoce kvalitní DNA ze 191 vzorků samičích květenství konopí, coţ je jedna z prvních úspěšně provedených izolací z květu Cannabis vůbec. V mnoţině 178 vzorků konopí bylo 72 SNP pozic detekováno metodou KASP v rámci technologie SNPline a účinnost detekce metodou SNPline byla 79 %. Bylo popsáno 57 SNP v neanotovaných oblastech genomu, které měly charakter tranzicí (51 %) a transverzí (26 %). Z 57 markerů navrţených na sekvenci v databázi bylo 44 jednonukleotidových pozic (77 %) polymorfních, a zbývajících 13 pozic (23 %) bylo v analyzované populaci monomorfních.

Abstract:

This thesis aims to shed light on the issue of classification and definition of two putative taxa of Cannabis, sativa and indica. The study focuses on assessing the variability between cultivars of Cannabis obtained as typical representatives of the sativa and indica types. Initial hypothesis was that chemical and genetic parameters are positively related to the two putative subspecies sativa and indica. The hypothesis was tested by chemical and genetic profiling, in particular gas chromatography and SNP analysis using KASP platform from LGC Genomics. Chemical analysis was performed using GC-MS with a set of 191 Cannabis accessions. Using this approach, 21 monoterpenes, 19 sesquiterpenes and 8 cannabinoids have been identified in total. From these, 8 of the identified terpenes were found in Cannabis for the very first time. The multivariate Principal Component Analysis elucidated the significance of terpenes when assigning samples from the supplier to either the sativa or indica type. Candidate terpenes were suggested as the potential biochemical markers for the distinction between these two types in the future. In this study, also high-quality DNA from 191 samples of female inflorescence of Cannabis using classical CTAB protocol was isolated, being one of the first successful isolations from the Cannabis flos ever. Out of the set of 178 samples, 72 SNPs were analyzed by KASP using the SNPline technology, the conversion rate being 79 %. Overall, 57 SNPs were identified in the nonannotated genomic regions, which had the character of transitions (51%) or transversions (26 %). Out of these 57 markers labeled based on genomic reads of Cannabis cultivars in database, 44 SNPs (77 %) were polymorphic, and the remaining 13 analysed positions (23 % of the tested population) turned out to be monomorphic.
