Regulation of beta-catenin signaling by receptor tyrosine kinases

Abstract:

In this thesis I study regulation of β-catenin signaling by tyrosine kinases receptors. β-catenin is a part of the axin, GSK3 and APC complex. It is present in cadherin complex, it is a part of Wnt signaling pathway. It has become subject of study because of its increased occurrence at various diseases. This work focuses on the role of β-catenin outside the Wnt pathway and looks at fosforylation of tyrosine residue 142, which leads to the dissociation α-catenin from β-catenin. It is this dissociation that leads to the disruption of cadherin/β-catenin complex and initiation of its transcriptional activity.

Abstract:

V této diplomové práci se věnuji studiu regulace β-kateninu pomocí tyrozín kinázových receptorů. β-katenín je součástí komplexu axin, GSK3 a APC. Katenin beta se nachází v kadherinovem komplexu, je součástí Wnt signální dráhy. Předmětem studia se stal pro jeho zvýšený výskyt při různých onemocněních. Tato práce je zaměřena na úkol β-kateninu mimo Wnt dráhu a soustředí se na fosforylaci tyrozinu 142, která vede k disociaci α-kateninu od β-kateninu. Právě tato disociace má za následek uvolnění β-kateninu z adhezivního kadherinového komplexu a iniciaci jeho transkripční aktivity.
