Abstract:

Transpozony jsou mobilní genetické elementy, vyznačující se svou schopností měnit pozici v rámci genomu. Vyskytují se ve všech genomech a často tvoří jejich podstatnou část. Svou aktivitou mohou způsobovat v genomu hostitele mutace, které mají většinou negativní účinek. Aby k tomu nedocházelo, bývají z převážné části umlčeny, zejména epigeneticky. Některé transpozony však mají schopnost reaktivace, která může být způsobena environmentálním stresem. Tento jev se projevuje hlavně u rostlin, které díky svému přisedlému způsobu života nemohou uniknout nepříznivým podmínkám a musejí na ně reagovat změnami genové exprese. Jejich genomy jsou pak plastičtější a náchylnější k přestavbám, na čemž se mohou podílet právě transpozony. Reaktivaci některých typů transpozonů pomocí abiotických i biotických stresorů se podařilo několikrát experimentálně prokázat u různých rostlinných organismů. Cílem mé diplomové práce bylo sledovat pomocí různých stresorů změny aktivity transpozonů Copia (LTR retrotranspozon), Ogre (LTR retrotranspozon), Chromogypsy (LTR retrotranspozon), SLACS (non-LTR retrotranspozon) a CACTA (DNA transpozon), které byly nedávno charakterizované v Laboratoři vývojové genetiky rostlin BFÚ AV ČR u dvoudomé rostliny Silene latifolia. K vystavení rostlin stresu jsem použila tři abiotické stresory: NaCl (jenž způsobuje osmotický stres), UV záření (stres ozářením) a CuSO4. 5H2O (oxidační stres). Ke sledování změn exprese transpozonů posloužila kvantitativní PCR v reálném čase s použitím relativní kvantifikace vztažené ke třem referenčním genům (aktin, 18S a EF1a). Obecně lze výsledky shrnout tak, že u osmotického stresu nedocházelo k výrazným změnám v expresi zkoumaných transpozonů. Stres ozářením způsoboval buď výraznou inhibici transpozonů, nebo nedocházelo ke změnám v expresi. U oxidativního stresu se projevila v listech mírná aktivace všech transpozonů. Výsledky se však jevily rozličně v závislosti na použitém referenčním genu a často se lišily i u opakování stejného experimentu. Z toho plyne, že sledování exprese transponovatelných elementů je velice náročný úkol, jenž je podmíněn mnoha faktory, které je třeba zohlednit. Je to zejména odlišná úroveň exprese transpozonů u jednotlivých rostlin a dále reakce referenčních genů na stres, kdy je potřeba použít správný referenční gen, jenž nebude reagovat na stres změnou exprese.

Abstract:

Transposones are mobile genetic elements, which posses an ability to change its location within the genome. They are occuring in all genomes, often constituting huge part of them. Transposones can cause mutations in host genome, which mostly end up with negative effect. Therefore they have been often silenced, mainly by epigenetic mechanisms. However, some of the transposones are able to reactivate themselves, what can be induced by environmental stress. This phenomenon is known to occur especially in plants, which as sessile organisms could not escape from adverse conditions and need to react by change of gene expression. Their genomes are more dynamic and prone to reconstruction, what can be mediated by transposones. Reactivation by abiotic and biotic factors have been experimentally reported for several types of transposones in various plant species. The aim of my diploma thesis was to observe how different stressors influence the activity of transposones Copia (LTR retrotransposon), Ogre (LTR retrotransposon), Chromogypsy (LTR retrotransposon), SLACS (non-LTR retrotransposon) a CACTA (DNA transposon), which have been recently characterized in Laboratory of Plant Developmental Biology IBP AS CR on dioecious plant Silene latifolia. For inducing stress in plants, three abiotic stressors were used: NaCl (causing osmotic stress), UV radiation (irradiation stress) and CuSO4. 5H2O (oxidative stress). A quantitative real-time PCR with relative quantification refered to three housekeeping genes (actin, 18S and EF1a) was used for monitoring transposone expression changes. In general, there were no significant changes in expression of selected transposones under the osmotic stress. Irradiation stress caused either strong inhibition of transposones or there were no changes in their expression pattern. On the other hand, oxidative stress slightly activated all of the transposones in leaves. However, the results are highly dependent on housekeeping gene used and often were different within two same experiments. From these obsevations follows that measuring the expression of transposable elements is very challenging task, which is controlled by multiple factors and all of them shloud be taken into account. Namely the different expression of transposones in plant individuals and reaction of housekeeping genes to stress, so it is necessary to use correct housekeeping gene, which will not respond to stress by changing its expression.
