Development and study of substances releasing carbon monoxide

Abstract:

Tato dizertační práce se zabývá studiem fotofyzikálních a fotochemických vlastností malých organických chromoforů, které po ozáření uvolňují oxid uhelnatý (CO) a vývojem molekul uvolňujících CO (photoCORM) po ozáření světlem z blízké infračervené oblasti spektra. Literární část předkládá předchozí publikované výsledky relevantní ke studovaným projektům, předkládá úvod do fotofyzikálních a fotochemických procesů, shrnuje výsledky výzkumu photoCORMů a jejich biologických aplikací, fotofyzikálních vlastností a možných fotoreakcí souvisejících sloučenin. Kapitola Výsledky a diskuze obsahuje dva na sobě nezávislé projekty. Cílem prvního projektu bylo studium fotofyzikálních a fotochemických vlastností derivátů kumarinu a studium mechanismu uvolňování CO po jejich excitaci. Bylo ukázáno, že látky podléhají komplexním vícekrokovým fotochemickým reakcím a že koncentrace kyslíku v roztoku a druh rozpouštědla mají důležitou roli v jednotlivých reakčních krocích. V druhém projektu jsem syntetizovala řadu nových photoCORMů obsahujících současně molekulu flavonolu a heptamethin cyaninu. V těchto látkách je molekula flavonolu zodpovědná za uvolnění CO a cyanin umožňuje absorpci spektra ve “fototerapeutickém okně”. Publikace a experimentální data jsou součástí přílohy v poslední části dizertační práce.

Abstract:

This dissertation is mainly about the study of photophysical and photochemical properties and mechanistic investigation of small organic chromophores that release carbon monoxide (CO) and the development of nearinfrared (NIR) light-triggered CO-releasing molecules (photoCORMs). The literature part provides the previously published results relevant to the studied projects, including the introduction of photophysical processes, the research state of the art of photoCORMs and their biological applications, photophysical properties, and possible photoreactions of relevant compounds. The results and discussion chapter covers two independent projects. The first project was to investigate the photophysical and photochemical properties of coumarin derivatives and study the CO release mechanisms. The compounds undergo complex multistep photochemical transformations. It was found that oxygen present in the solution and the type of solvent play a key role in the individual steps. In the second project, I worked on the synthesis of a series of new photoCORMs incorporating flavonol into the structure of heptamethine cyanines. In these photoCORMs, flavonol is responsible for the CO release, and cyanine provides the absorption shift to the phototherapeutic window. The publications and experimental data are appended to the end of the thesis.
