Dysregulation of microRNA in relapsed multiple myeloma patients

Abstract:

Mnohočetný myelom představuje druhé nejčastější hematoonkologické onemocnění charakterizované infiltrací kostní dřeně plazmatickými buňkami, které produkují monoklonální imunoglobulin. Především díky použití nových léčiv se významně zvýšila kvalita a délka života pacientů s tímto onemocněním. Mnohočetný myelom však zůstává stále obtížně léčitelný a téměř všichni pacienti relabují. Toto onemocnění se vyznačuje velkou heterogenitou, a proto u pacientů dochází k relapsu v odlišném časovém období. Zatím není možné předpovědět, za jak dlouho budou pacienti relabovat, avšak četné studie zkoumající dysregulaci nekódujících molekul RNA u nádorových onemocnění naznačují, že mikroRNA by mohly být dobrými markery relapsu. V této práci byly pomocí sekvenování nové generace hledány rozdílně exprimované mikroRNA mezi třemi skupinami pacientů s mnohočetným myelomem, kteří relabovali v různých časových intervalech. Poté byly validovány tři mikroRNA s největším rozdílem exprese mezi nejodlehlejšími skupinami a u jedné mikroRNA došlo k potvrzení signifikantně rozdílné expresi. Následně byly identifikovány rozdíly v hmotnostních spektrech mezi skupinami s použitím hmotnostní spektrometrie využívající laserovou desorpci a ionizaci za účasti matrice s analyzátorem doby letu.

Abstract:

Multiple myeloma represents the second most common haematological malignancy characterized by infiltration of the bone marrow with plasma cells that produce monoclonal immunoglobulin. Primarily thanks to the use of new drugs, the quality and length of life of patients with this disease has significantly increased. However, multiple myeloma remains difficult to treat, and almost all patients relapse. The disease is characterized by a high degree of heterogeneity; therefore, patients relapse at different times. It is currently not possible to predict when patients will relapse, but numerous studies investigating the dysregulation of non-coding RNA molecules in cancer suggest that microRNAs could be good markers of relapse. In this study, differentially expressed microRNAs among three groups of multiple myeloma patients who relapsed at different time intervals were analysed using next-generation sequencing. Three microRNAs with the largest difference in expression between the most distant groups were validated, and significantly different expression was confirmed for one microRNA. Subsequently, differences in the mass spectra between groups were identified using matrix-assisted laser desorption/ionization time of flight mass spectrometry.
